To begin with, simple inspirations can explain a lot about the character's mysterious motives, especially the inspirations that the author had with certain films, specifically "Human Centipede".
This film is referenced twice, being mentioned through Junpei's conversation with Yuji and in the anime Ep 6 of the Second Season by Yuji. The two human centipede films talk about a mad scientist wanting to experiment on people.
Obviously alluding to Itadori being an experiment by Kenjaku, even the baby worm's collar is the same as Yuji's red collar.
Junpei even quotes a phrase about the mad scientist from the film Human Worm 3: where changes from the perfectionist who abandon absolutely everything occur. This creates several questions, in the past did Kenjaku abandon his dream of creating the perfect being?
This is said when Junpei and Yuji were talking about movie three, in season two, Yuji wanted to see movie 4, where we see the opposite of perfection, a monster, something we can consider Twisted?

Based on all this, we can then understand that Kenjaku was trying to bring Sukuna back by creating the only Vessel that would support the King of Curses, Sukuna's twin brother. It took ages of experimentation for this to occur.
