About the vaccine in the making,My views 👇
University of Oxford/AstraZeneca,CanSino Biological Inc./Beijing Institute of Biotechnology, Gamaleya Research Institute(Russia) these are into Non-Replicating Viral Vector(Adenoviral).1+
This will be the first human vaccine using adenovirus as a vector. There are many animal adenoviral vector vaccine. Why not in human b4 ? coz this vector has many serotypes cause many types of diseases from eyes to GI tract. So not used before.2+
Recent Adenoviral vector is the attenuated one,modified non replicating type so safe ,may be.This vector has Spike glycoprotein. So after injecting the cells will express S protein of virus &will stimulate immune response.3+
Sinovac,Wuhan Institute of Biological Products/Sinopharm, Beijing Institute of Biological Products/Sinopharm all these uses inactivated/killed virus. Since it is the whole virus that is killed,effective doubtful?? Long lasting immune response Doubtful??specificity questionable.4+
Coz this RBD epitope is not targeted.
Moderna/NIAID, BioNTech/Fosun Pharma/Pfizeris on to mRNA as a vaccine.This is a new generation vaccine.here mRNA is given ,our cell ribosome will translate it and produce specific protein and express it.5+
New,yet seems promising,due to specificity.Chinese vaccine may not be reliable,inactivated vaccine is a conventional vaccine, how it is inactivated we have to see that also,chemicaly or by other method.Transperancy needed to be secure.6+
Russian and astrazena vaccine after proper trial may be more reliable.7/7🙏🙏😃
About mRNA Vaccine delivery system,
Since it a new type of vaccine, many of us are unknown about it.
mRNA delivery is a critical process. Exogenous mRNA must penetrate the lipid membrane barrier & reach the cytoplasm & be translated to functional protein. 8+
mRNA uptake mechanisms is cell type dependent & the physicochemical properties of the mRNA complexes can influence cellular delivery.
2 approaches for the delivery of mRNA vaccines are generally applied.9+
1.Loading mRNA directly in dendritic cells exvivo ,then reinfusion(allows precise control of the cellular target).2. direct parenteral injection of mRNA with or without a carrier(rapid,cost-effective, but it does not allow precise & efficient cell-type-specific delivery).10+
preferentially targeting antigen-presenting cells, by intradermal & intranodal injection or in tissues using a gene gun, a microprojectile method.11/11🙏😄
we should not forget that naked mRNA is quickly degraded by extracellular RNases & is not internalized efficiently, different transfection reagents are used to facilitate cellular uptake of mRNA &protect it from degradation.12/12🙏😄
