Fronto-temporal lobar degeneration with TDP-43 (FTLD-TDP) is a fatal neurodegeneration. TMEM106B variants are linked to FTLD-TDP risk.
TDP-43 is a common substrate for SarsCoV2 Nsp5. Nsp5 appears to be cytotoxic towards neuroblasts and glial cells demonstrating a direct killing effect of SarsCoV2 infection on neuronal cells through TDP-43 cleavage, which may lead to further CNS dysfunction.
Nsp5 is a viral neurotoxic factor, and its 3CLpro activity is a druggable target for relieving neural damage.
