Some vehicle is needed because DNA/RNA have the same charge as a cell membrane, making them repulse each other.
Early ideas, like using a cationic polymer (polymers that are positively charged) worked but have short-comings when it comes to transfecting live whole animals (like humans).
Instead Cationic Lipids/Liposomes were developed/designed to overcome those problems.
It is still not clear how exactly a DNA payload crosses the nuclear envelope, but they mostly understand how things get into the cytoplasm.
Transaction Reagents have been created that work w/ a wide variety of eukaryotic cell lines. (Meaning they work w/ cells that are not bacteria β for ex β & are traditionally more resistant to taking in foreign material.)
Side thought: Why do bacteria readily take in material?
Nonliposomal methods have also been created.
Side Note: Cullis? Pieter Cullis? π€
The original transfection methods were Physical Methods including microinjection, electroporation and particle bombardment (the "pew pew" method).
It's interesting how in the shiny product brochure they reiterate several times how various transfection reagents have "low toxicity", but then emphasize that something has to be done about the toxicity of the reagent. π€π€π€
Just a side note in most biological measurements, you NORMALLY need a "reference" measurement. Bc many things are changing, how does the researcher know if the expression of what they're looking for is going up or down when everything is going up?
Yes, looking at you PCR tests.
So in the end, transfections are not new. They are a product that has been used in experimentation for a long time. They were classically used for research needing gene transfers, but have also been used for a variety of different things. Most bench biologists would know this.

@jjcouey presents to Dr. Sucharit Bhakdi on Medical Doctors for COVID Ethics International yesterday (Oct. 17th) hosted by @CharlesKovess with guest appearances/discussions by @NickHudsonCT, @SenRonJohnson and @davidrasnick1.
(Transcripts available...)
If @CharlesKovess hasn't added the transcripts to the Rumble stream yet, you can "hack" around this using this jsfiddler page I put together.
It overlays the diarized transcriptions on the video
CSASPP July 29, 2024 update has some interesting thinking on Bonnie Henry's recission.
(Unfortunately this site doesn't have anchors so I can't send you directly to the entry. Instead just search for "29 July, 2024:" in your browser.) covidconstitutionalchallengebc.ca/status-updates
Kip's theory is that this is a business decision based on potential future liability increases. His observations that none of our actions or advocations had any DIRECT effect is probably correct.
However, despite this, the court of public opinion DOES indeed affect the courts. But understanding the mechanism at which things change is useful in how we direct our efforts.
Understanding the @IamBrookJackson case by @barnes_law from the start. (1.35x spd)
"... surely they'll step in. They realize it's a mistake. ... Then she got word back ..., well, we really can't do anything because this is happening at all the clinical trial testing sites."

"Suddenly, she's summarily fired. She reaches out and blows the whistle to the FDA anonymously, confidentially. That same afternoon, she gets a call from Pfizer's counsel on a phone number that only the FDA's confidential, secure whistleblower line knew, ..."

"..., no intention of ever prosecuting the case. I suspect they've been deliberately sabotaging your story and delaying it as long as possible before you could do it, before your information could be used to stop what they're doing."
"... redefine democracy from being about the will of the voters to being about the sanctity of democratic institutions. & who are the democratic institutions? ..., it's the military, it's NATO, it's the IMF and the World Bank. It's the mainstream media, ..."
"At the end of the day, a bunch of, you know, populist groups decide that they like a truck driver who's popular on TikTok more than the carefully constructed consensus of the NATO military brass. Well, then, from their perspective, you know, that is now an attack on democracy."
From ~47min+.
Basically, our democracy is different from their democracy. That is how the real world works now. Until we understand this and try to do something about it, nothing will change. They don't want us to do anything about it.
π€ A while back @barnes_law claimed @sasha_latypova was wrong about the fact military contractors were involved and that if it was true then the @IamBrookJackson case had no teeth because you can't sue the government. Not sure what this means now. I'll see if I can find it again.
Before I get continue, there's been a slew of π going on recently. This isn't one of those.
I am trying to keep track of what's going on & to better understand. I tend to forget details over time (small π§ ). These are my own notes which I'm sharing.
The Stiftung Corona-Ausschuss (Corona Investigation Committee) with @wodarg talks to @HousatonicLive who reveals how the drug epidemic may have contributed to the illusion of a pandemic. It also touched base with Lyme Disease.
Session 194 : Mark Kulacz
There's definitely some very interesting discussion to be heard.
- >1/2M deaths in the US (2019-2023) were opioid ODs! (for a country of 330M)
- for 2023, 1:30 who died were drug ODs!
- comparatively in 2000, OD deaths were ~22K, vs ~113K in 2023
