The results show that gastruloids are organized and that this organization corresponds to that of the embryo; see go.nature.com/3d0lB6b for details.
One surprising observation here is the differentiation trajectories of NMPs and, in particular the gene expression sequence of somitogenesis: Bra ->Tbx6 -> Uncx4.1.

#BSDBGenSoc2020 These days we all have other things in our minds, it is difficult to concentrate and many of my thoughts R w/ family anf friends in #Spain) but discussing our work, is an important part of our being and it is going to be important in the weeks and months to come.
While many, all, of us rather do al this in person, we are going to have to adjust in the weeks to come. #BSDBGenSoc2020 is a good way to explore how we are going to do it so, THANKS @_BSDB_ and @the_Node for organizing and running this.

Today I was meant to be presenting at #BSDBGenSoc2020 but rather, I am preparing to close our lab. However, I am able to share the poster and talk I was meant to be giving today as part of the Plasticity in Developmental Genetics session. Image
We investigated the requirements of yolk derived signalling and geometry on pattern formation. By explanting the whole zebrafish embryo at the 256 cell stage and cultured them in a basic L15 3% FBS media. The explants elongate after 6 hours, movie from @viktri08 @andattardi
At the protruding end, we observe expression of the mesodermal marker Tbxta (brachyry), Tbx16, Protocaderin8 and Noto Image

@the_Node @SCICambridge 1. We hypothesised that two chromatin remodellers with apparently opposing biochemical activities would antagonistically maintain enhancer structure. Loss of either should have opposite conseqeunces for gene expression, transcription factor binding, and response to signalling.
@the_Node @SCICambridge 2. Using inducible degradation systems and careful temporal analysis, we proved that this is WRONG!
