Bottom-line-up-front: stain overnight. Antibody staining is sensitive to both dilution and time, so a titration that gives crummy staining in 30' can actually give beautiful staining overnight. The best part is, that lower dilution gives less background, for better separation 2/6 Image
You can see this quite clearly with even tricky staining, like #Foxp3 and #cytokines. The overnight stain gives a stronger positive signal without increasing the background at all. Plus your cells are ready in the morning, when the cytometer is free, rather than 8pm at night! 3/6 Image

We tested >12,000 genes to find positive regulators of T cell proliferation to be used for next-gen #immunotherapies.

(2/28) T cell therapies have shown the potential to cure patients from #cancer – but even in B cell cancers, relapses outnumber cures… One of the problems is limited T cell persistence. #celltherapy #CARTcell
(3/28) We wanted to find new genes (including genes never expressed in T cells) that could improve T cell persistence. Now, the question was: should we use #crispr activation or directly deliver target genes on a #lentivirus?

Congratulations, Agnieszka @BartenschlagLab! We actually began working on this during the last pandemic threat (#Zikavirus) and the way through editor and reviewer offices was not exactly aided by the current pandemic. But now it’s finally out! jvi.asm.org/content/early/…
Through TLR3, ZIKV induces proinflammatory #cytokines (IL-6, IL-10), which trigger STAT3 signaling, which in turn dampen #interferon responses. As #RIGI and #MDA5 sense ZIKV and induce IFN production, this antagonism eventually aids viral replication as we can show.
This is all so weirdly complex– a lot of work ahead to understand the details of innate immune regulation in virus infection. Ha, saying that– if you made it all the way to here in this thread, consider contributing to this special issue on exactly this topic ⬇️

The immune system protects the host from invading pathogens through 2 layers: the innate and adaptive immunity. Innate immunity acts within minutes to remove the pathogen. Adaptive immunity takes days/wks but is specific & long lived, and is the basis for #vaccines. (2/n)
Innate immunity starts by triggering sensors that detect unusual molecules or activities, known as pattern recognition. Once microbial patterns are detected by these sensors (#PRR), they trigger immediate alarm through secretion of #cytokines and #interferons. (3/n)

A new paper in @sciencemagazine from #Belkaid lab at #NIH described an #alternative differentiation #pathway for #commensal-specific T cells in presence of inflammation. Do the data presented support conclusions? Let's dive in.

1. #Commnesal S. epidermidis–specific -#MIIINA:H2-M3+ CD8+ T cells ordinarily differentiate into Tc1 or Tc17 subsets. However, when co-exposed to chitin or sand fly (source of #inflammation), Tc17 subset also produce type II cytokines.
2. In such condition, type II #cytokines such as IL-5 or IL-13 are produced by CCR6+ Tc17 cells (out of which 30% are IL-17+). However, there is no overlap between IL-17+ and IL-5+/IL-13+ CD8+ T cells. So, the authors conclusion about "paradoxical phenotype" is unsubstantiated.
