#Spike protein’ is the major target of #NAbs in infected/vaccinated people. The prefusion conformation of the Spike represents the most relevant target because Abs can successfully interfere w/ infection only if they prevent binding to target cells or prevent fusion itself 1/
Various approaches are adopted to express ‘#Spike’ protein in vivo, such as nucleic acid structures & virus vectors. Alternatively, other vaccine platforms like recombinant protein particles & inactivated vaccines can directly deliver this protein 2/
Most structural studies of the wild-type spike protein exhibited one #RBD in the 'open' conformation. By contrast, 2 or 3 RBDs were observed in the open conformation in the more infective #D614G mutant--the RBD #conformation plays a vital role in the infectivity of SARSCoV2 3/

Is there any role of large-scale vaccination in containing the ongoing surge in #India? The current estimate of herd immunity threshold is around 60%-70% based on the estimate with old variant #D614G. 1/
With the advent of new, more transmissible #variants, the estimate for a herd immunity threshold may further go up. The pace of inoculation is very sluggish, inoculated a meagre 2.0 % of our population with 2 doses! May take few years to reach anywhere near the threshold 2/
We are currently on the ascending limb of an outbreak propelled by a highly transmissible virus with a high reproduction number (Ro). Further, there is confusion over what it means to reach herd immunity 3/

(2) Four sets of Hamsters were infected with either mock virus, D614 variant, G614 variant and a 50/50 mix of both variants of the coronavirus.

Hamsters harvested for tissue analysis on day 2, 4, and 7 of a 7 day study.
(3) Trachea, Lung Sections and Nasal Wash were collected and analyzed to measure the concentration of the two variants.

(2) A group of researchers prepublished their findings about the #D614G mutation to the SARS-Cov-2 affecting the properties of the Spike protein that binds to host cell sites.
(3) The paper includes a diagram showing the emergence of the #D614G mutant and its descendants, first in Belgium, and later in hot spots in Europe and the United States.

The relative quantities of the this mutant strain could explain the variations seen in Case Fatality Rates!

🚨US scientists say a new #coronavirus strain emerged in March and now dominates worldwide. The mutant (spike protein #D614G) appears to be *more contagious* than versions that spread early in the #CoronavirusPandemic and may bypass immunity to 1st strain.
latimes.com/california/sto…
Wherever the new #coronavirus strain appeared, it quickly infected far more people than earlier strains from #Wuhan, and within weeks it was the ONLY strain dominant in some nations. The new strain’s dominance demonstrates that it is more infectious, but it's not yet known why.😳
Prior scientific reports did NOT reveal any significant mutations that affected the infectivity of the #coronavirus. This new report is very troubling, as it may mean that immunity to the 1st strain or vaccines directed against the 1st strain may NOT be effective against the 2nd.
