Glioblastoma (GBM) is one of the deadliest, most aggressive cancers that exist, with a median survival of only 15 months.

In GBM, 'single cell heterogeneity' are not simply buzzwords.

Rather, this immense heterogeneity is a main reason of treatment failure Image
Recent work demonstrated that single GBM tumors are mosaics of cells in different states, each associated with distinct genomic driver alterations.

While transitions between cell states can occur, each genetic driver favors a particular cell state.

Question that hit me on October 26th, 2017: is there a temperature window that stops 🛑 tumor growth, and the brain 🧠 can tolerate? If so, this could prevent tumor recurrence and buy the patient time to try therapies until we reach a cure.

1. Tumor cells stop growing at room temperature (20-25°C). Remember, body temperature is 37°C.
Even 18 hours of cooling per day can be enough! 🕒 This point will be useful for a device in the future.

Background: Standard of care (SOC) and patient survival in glioblastoma have changed little in the past 17 years.
We evaluated in a phase 3 trial whether adding an autologous tumor lysate-loaded dendritic cell vaccine (murcidencel) to SOC extends survival. Patients and

Methods: Newly diagnosed glioblastoma patients were randomized 2:1 to either murcidencel or placebo.
Under a crossover design, all patients could receive murcidencel following tumor recurrence. All parties remained blinded regarding treatments before recurrence. Patients thus received murcidencel at new diagnosis (nGBM) or at recurrence (rGBM) following crossover from placebo.

Some quick thoughts on $NWBO and #DCVaxL. DCVaxL is designated an #OrphanDrug, for an #OrphanDisease, #Glioblastoma. This provides a bunch of benefits which are off topic for this thread.
My main point is to suggest that patients needing #DCVaxL may likely qualify for either of two funds in the UK, the #InnovativeMedicinesFund or the #CancerDrugsFund, which together have £640M allocated to provide early coverage to patients in the UK's NHS.
The UK charity, BrainTumorResearch.org recently put out a PR about the new program in the UK for rare diseases, which is modeled on an already existing program called the #CancerDrugsFund.

2/ $NWBO and UCLA have a patent application pending approval for immunotherapy combinations with #DCVaxL.
3/ The #DCVaxLComboTrial appears to be intended to further the patent process and also to likely advance new commercial opportunities, including potential partnerships, for $NWBO. Many commentators have suggested some obvious potential partners.

Good morning. Not looking for sympathy. Sharing the truth hoping to raise awareness.
Please remember Glioblastoma multiforme is a grade IV brain tumour. I am Not in remission. There is No Cure. I still have piece of the glioma in my head.
This unique form of brain cancer and the survivorship is extremely grim. The Life expectancy is 12-16 months and the five-year survival rate is only 6.8 percent.

I get inspired by real world problems that I feel are solvable with data, because then it becomes within my scope to tackle. If I look at #glioblastoma I see room for #blockchain to provide clarity on #patientconsent for uses to enable monetization, ideally to help offset costs.
To unpack these things further, I mean leveraging smart contracts for dynamic consent or 'prosent' as presented here: jme.bmj.com/content/early/…
This would allow for the patient to opt into selling their data to researches, insurance or pharmaceutical companies

Very pleased the Senate passed our resolution designating July 22, 2020 as #GlioblastomaAwarenessDay.

Glioblastoma is a devastating brain disease that has taken the lives of two of my very close friends and colleagues, Ted Kennedy and my dearest friend John McCain.

More on immune evasion in glioblastoma! Online now in @NatureMedicine, intracranial tumors drive sequestration of T cells in the bone marrow through downregulation of S1PR1 receptor surface expression. nature.com/articles/s4159…
GBM patients, including treatment-naive, and mice with a variety of intracranial tumors (regardless of tumor origin) show extremely low levels of circulating T cells and reduced spleen size, with accompanying increase in bone marrow T cells (especially naive T cells).
This seems to be due to lower S1PR1 on T cells, driven by ligand-independent internalization (RNA levels are identical between control and tumor-bearing mice and there is no increase of S1P in the tumor-bearing mice).

Reading this review on immunotherapy in glioblastoma (nature.com/articles/s4157…) @NatRevClinOncol . A few odds and ends make me wonder - is there going to be a special role for NK cells in glioblastoma? Should we be aiming to target them for more effective therapies?
Checkpoint blockade hasn’t shown much efficacy in glioblastoma, in line with relatively low mutational burden (nejm.org/doi/full/10.10…) @NEJM. Also in line with the brain as “immunoprivileged”, or at least as a place with "unusual" immune activity.
First started thinking about NK cells in GBM after reading excellent study on PDGF-DD as an activating NK cell ligand (sciencedirect.com/science/articl…) @CellCellPress - striking human data from GBM cohorts, especially with greater infiltration of NK cells than T cells into tumor.
