#location location location: thrilled to share our work @CellCellPress! #interferons dictate #COVID19 severity! Specific #IFNs are produced along the #airways in response to #SARSCoV2 and drive protective or detrimental #transcriptional programs! 1/7 doi.org/10.1016/j.cell…
IFNs are driven by high #viral #loads, suggesting that viral recognition is only achieved when #SARSCoV2 reaches a certain threshold! Efficient induction of protective IFNs is preferentially achieved by younger patients, further explaining susceptibility of elders to #COVID19 2/7
#IFNL1 & #IFNL3 characterize mild illness & drive potent anti-#SARSCoV2 #ISGs in the upper #airways. Severe patients show #typeI IFN & #IFNL2 along the respiratory tract but lack ISGs, dampen cell proliferation programs associated to #repair and boost #apoptotic #p53 pathways 3/7

Developing an #mRNA vaccine is a sure-shot way to success? No! Despite the spectacular performance of #Pfizer & #Moderna vaccines, we also have a #CureVac fiasco. We need to dig in much deeper into the intricacies of developing a highly efficacious mRNA vaccine 1/
There is more than low dose & impact of VOCs behind #CureVac’s failure. And here we once again need to salute the sheer brilliance of @kkariko . Will do a little thread to explain, why! 2/
No doubt, the #CureVac used a much lower #RNA dose. Dose is lower (12 (CureVac) vs 30 (#Pfizer) vs 100 ug (#Moderna)) but no idea how that translates to protein. Need to know how much of a role the lower dose played in the lower efficacy 3/

Our latest study to understand the dynamic regulation of #interferon (#IFN) and #SARSCoV2 infection.

🧵summarizing what we found and what needs to be done to understand the dynamic interplay of infection and #IFN regulation. @iScience_CP #COVID19

We infected human airway cells (Calu3) with #SARSCoV2, and performed a time-series #RNAseq analysis to discover global transcriptional responses. We saw expression of #IFN (beta and lambda) and IFN stimulated genes in these cells.
Here is what #SARSCoV2 infection looks like in #Calu3 (human lung epithelial cells) cells.

Very excited to share this work by the heroic team - @carolilucas Pat Wong @sneakyvirus @tiago21bio et al on “Longitudinal analyses reveal immunological misfiring in severe COVID-19”. A thread on our findings. #COVID19 #Pathogenesis (1/n)

In this study, we enrolled COVID+ patients and analyzed their immune responses and viral load over the course of their hospital stay. We also compared samples from COVID- healthy health care workers (HCWs) as comparison. (2/n)
As reported by others, we found that COVID patients with severe disease have low T cell numbers and increased monocytes and neutrophils. We also found eosinophils come up in patients. This is bizarre 🧐 (3/n)
