It all started when we wrote something with @palomi_rock
The intro was so good & had so much new data that we decided to make a 'short' review/update of pannexins in #immunecells, focus on cell migration.
But things got out of hand & we started to include more cell types 😅🤷🏽‍♂️

Neutrophils are unique because they can use their chromatin as a weapon to fight pathogens in a process termed 'NETosis'. They are extremely short-lived, surviving only ~6h after terminal differentiation, and have thus not been extensively studied throughout organismal aging. 2/n

These are diverse diseases & increasing age technically not a disease at all. However, there is at least 1 factor in common among all of the co-morbidities identified thus far:

INFLAMMATION

A survey of the co-morbidities revealed another commonality; a close association w/ dysregulation of the pro-inflammatory, neutrophil-recruiting cytokine, IL-17A.

IL-17A is made by multiple immune cell types, including lung-resident innate lymphoid cells (ILC3s).

Circulating neutrophil precursors, aged neutrophils, and a few populations of mature and immature neutrophils. Melanoma stage correlates with loss of dominant N2 (mature, ROS++, lowly phagocytic), and increase of N5 (immature, non-proliferative, ROS+, lowly phagocytic).
Interesting to note the phagocytosis-SSC staining in Fig4A - big changes in SSC for some populations with zymosan, others not so much - degranulation, different phagosomes…? Similarly, bimodal peaks for ROS production in same populations… Image
