It turns out that the positional information they convey to #cells in the #neuraltube is much more accurate than previously estimated!

A methodological subtlety has previously led to overestimated positional errors of the morphogen gradients in the mouse neural tube. We reanalysed the noisy gradients and found that the positional information they can convey to cells is actually orders of magnitude more precise. Image

This is the most straightforward explanation for chronic symptoms in at least a subset of #LongCovid patients. Partly b/c if the #virus is still present its activity can directly contribute to other phenomena also being documented in LongCovid

3/ Persistence of SARS-CoV-2 can lead to ongoing downregulation of interferon and/or T cell signaling by the virus, creating an optimal atmosphere for activation of EBV or other #pathogens normally controlled by such immunity

New preprint: The team did autopsies on 44 patients w/ #COVID-19 to map + quantify SARS-CoV-2 distribution, replication and cell-type specificity across the human body (including brain) from acute infection through over 7 months following symptom onset: researchsquare.com/article/rs-113…
2/ They found that #SARS-CoV-2 was widely distributed, even among patients who died with asymptomatic to mild COVID-19. They also detected persistent SARS-CoV-2 RNA in multiple anatomic sites, including regions throughout the #brain, for up to 230 days following symptom onset
3/ Persistence of low-level #SARS-CoV-2 RNA was frequently detected across multiple #tissue categories among all late cases patients who passed away after 31 days, despite being undetectable in plasma

Important paper 👉 A team in Tokyo took RNA-seq data from the Genomic-#Tissue Expression Project: a public resource created to study tissue-specific gene expression/regulation from 54 tissue types collected from 1000+ healthy individuals at autopsy: bmcbiol.biomedcentral.com/articles/10.11…
2/ They successfully identified 39 viral species in at least one tissue (tissue types included #brain, pituitary, esophagus, thyroid, #heart, breast, lung, kidney, adrenal gland, prostate, #nerve, adipose tissue, blood vessel, ovary, uterus etc)
3/ Viruses identified in the various tissue samples included EBV, HSV-1, Varicella, CMV, HHV6-A/B, HHV-7, HCV, HPV, adeno-associated virus...and 16 RNA #viruses including Respiratory syncytial virus (RSV), Parainfluenza Virus 3..

Thanks for sharing my perspective in this piece @MeganEDoherty! As I mention, in these #longcovid cases, it’s very important to take the potential persistence of #COVID-19 in certain tissues and/or the central nervous system (CNS) seriously!
2/ That’s b/c #Coronaviruses have been shown capable of persisting in human #tissue or certain “anotomical sanctuaries” - for example, this team found Coronavirus RNA + antigen in the Multiple Sclerosis brain: ncbi.nlm.nih.gov/pmc/articles/P…
3/ Also in other “post-viral” syndromes like “Post-#Ebola Syndrome, viral RNA has been found in unexpected body sites like sperm, or the eyes, years after initial infection: nejm.org/doi/full/10.10…

Cool new study which found that a range of #pathogens (some capable of long-term persistence in tissue + #blood) upregulate expression of the cell surface protein CD47 in the host cells they infect as an #immune evasion strategy
2/ I fact, upregulation of CD47 is a very smart survival strategy by such pathogens, b/c it interferes w/ the host innate immune response that is normally supposed to identify, target and kill them!
3/ Specifically the team found that upregulation of CD47 by the pathogens under study slowed uptake of dead/#infected cells by the #immune system, including the downstream activity of #antigen presenting cells that are needed to recognize pathogens in the first place
