It’s been awesome watching this story develop over the last couple years. Truly fantastic effort by the whole group on this intriguing patient cohort, complete with some exciting and new longitudinal data on stability of the microbiota in IgA-deficient patients.
I’ll let the story speak for itself (go read it!) - important work on human immunology and the role of IgA in shaping the gut microbiota. Way to go Jason!

The bottom line: Best RNP electroporation conditions resulted in up to 80% deletion, consistently >50%, of 3 tested genes for activated primary human NK cells. Validation of Ncr1 and Cish biology from mouse studies in human NK cells.
Paper starts by demonstrating issues with lentiviral transduction in human NK cells. Electroporation was optimized very thoroughly (>10 buffers and pulse conditions), with ~doubling in efficiency (viability and uptake) to >80% transformed and >80% viable

Very cool pair of new preprints @biorxivpreprints from @barton_lab on TLR regulation by Unc93b1 - biorxiv.org/content/early/… and biorxiv.org/content/early/… Here's my summary and a couple thoughts... #TLR #CellBiology #Immunology #PreReview #ASAPBio
First one, on TLR9 regulation by Unc93b1. Triple-alanine scanning mutagenesis library revealed mutant of Unc93b1 (SKN) that abolishes TLR9 signaling, although leaves endosomal trafficking and maturation intact. Single serine mutant (S282A) is responsible for full SKN phenotype
SKN or S282A both reduce amount of TLR9 binding to CpG. Both also increase amount of TLR9 bound by Unc93b1. Other mutations in same region of Unc93b1 (loop 5) either enhance or block Unc93b1 binding but all impair TLR9 activation, with much less effect on TLR7

Very cool paper on species-specific fertilization in fish mediated by a single Ly6/uPAR protein, Bouncer. biorxiv.org/content/early/…
Bouncer expressed on oocytes is necessary for fertilization in zebrafish by mediating sperm-egg binding and entry. And, incredibly, oocytes expressing Bouncer from a distantly related fish (medaka) could be fertilized by medeka sperm, but not zebrafish sperm, and form chimeras!
This is super interesting, and the molecular mechanisms will be very interesting to follow up. In particular, what's the sperm receptor for Bouncer (since Bouncer is only required on the oocyte - it's not a homotypic interaction)? How did that pair of proteins co-evolve?

Destabilization of NLRP1b (e.g. cleavage by bacterial protease or degradation by bacterial ubiquitin ligase) releases non-covalently linked C-terminal fragment that can activate inflammasomes - caspase-1, pyroptosis, IL-1b, the works. (2/9)
Awesome mechanism for maintaining auto inhibition with associated fragments after auto-catalytic cleavage, and exploiting processive nature of the proteasome - using Nlrp1b as a “decoy sensor” sensing its own stability, a molecular "booby trap". (3/9)
