Can a respiratory #virus infection confer #immunity against the other #respiratory virus? A much-debated issue in the virus immunology for many decades. What is the probable immune mechanism & does it indeed exist? What is the evidence for & against this hypothesis? 1/
It is hypothesized that a respiratory virus infection confers immunity against the same and other respiratory viruses for a short time, perhaps a few weeks. This immunologic mechanism, known as #heterosubtypic ‘temporary non-specific immunity’ 2/
This immune protection is associated with activation of the innate immune response to viral infection mediated by the release of Type I #interferons (IFN) & other cytokines that have broad protective effects against a range of viruses 3/

Our latest study to understand the dynamic regulation of #interferon (#IFN) and #SARSCoV2 infection.

🧵summarizing what we found and what needs to be done to understand the dynamic interplay of infection and #IFN regulation. @iScience_CP #COVID19

We infected human airway cells (Calu3) with #SARSCoV2, and performed a time-series #RNAseq analysis to discover global transcriptional responses. We saw expression of #IFN (beta and lambda) and IFN stimulated genes in these cells.
Here is what #SARSCoV2 infection looks like in #Calu3 (human lung epithelial cells) cells.

In this study, we have developed a fast and cost-effective method
for reverse genetics of coronaviruses by combining two approaches developed by others..As the genomes can be divided into multiple short fragments, mutations can be introduced into individual
fragments easily. ImageImageImageImage

As the elderly are more susceptible to #COVID19 w/ signs of lung #inflammation, here's a thread about our @virusesimmunity 2016 flu paper on #antiviral resistance, excessive lung #inflammation, & a strategy to boost disease tolerance! science.sciencemag.org/content/352/62…
First we demonstrated that monos & macs from older adults have impaired type I IFN (antiviral) & #ISG responses but intact cytokine responses after #flu infection. To probe the possible in vivo consequences of weak #IFN responses in the face of robust inflammation...
... we made a mouse model that mimicked this immune response in the elderly (Mx1+/+ mice w/ genetic deletion of tlr7, mavs). Failure to induce type I IFN led to high viral titer, 2º bacterial infection, severe lung inflammation, and death.
