1/🧵We developed new in vivo #ILC models injecting intraductally ER+ #ILC lines and freshly dissected ER+ ILC tumors @EmboMolMed embopress.org/doi/full/10.15…
2/🧵#ILC cells establish themselves at the ductal tips #microenvironment and form grape-like structures reminiscent of #TDLUs of the human breast.
3/🧵We show that in these models, the tumor cells grow slowly, invade forming hILC histologies, and metastasize in a similar way as they do in patients in clinical relevant organs including ovaries, brain-meninges, adrenal gland, GI tract, liver, and lung.

In (breast) cancer research we use a few different model systems to study cancer growth and development:
1) Cell culture
2) Animal models
3) Tissue analysis
4) Ex vivo models
Cell culture is a method to study cells in a petri dish. The cells are taken from a person's tumor, media is added, and the cells are allowed to grow. We grow them in an incubator so the conditions are similar to the body.

This picture is cells called BT474. 📷: @officialatcc
The media we add contains all the nutrients that cells need to survive for baseline growth including amino acids, glucose, and growth factors.

We place the cells into smaller wells to treat them with different drug treatments or hormones then analyze what happened to the cells.

When someone is diagnosed with cancer, part of the tissue is removed and put into a solution that fixes it in time and place (called a fixative).

The tissue is then cut into very thin sections, put on a slide, and stained with dyes that help pathologists to visualize the tissue.
I took an amazing histology and pathology course when I was doing my PhD at @RutgersU in New Jersey USA. (Hi Dr. Reuhl!)

The first and most common stain is hematoxylin & eosin (H&E). Here are two pictures of normal skin and cancerous skin. Can you tell the difference?
Hematoxylin stains nuclei dark blue or purple while eosin stains proteins in the cytoplasm of cells. While it’s great to show structure it doesn’t distinguish cells from connective tissue.

Enter: trichrome stain.

red= keratin and muscle fibers
blue= collagen
orange= RBC

Clinical trials design for #lobular #breastcancer patients are finally available! Let's look for a moment at the Pitt Neoadjuvant Endocrine therapy trial.
lobularbreastcancer.org/clinical-trial…
#bcsm 1/
In this trial, patients will be treated before surgery for 21 days with tamoxifen, anastrozole, or fulvestrant (all current drugs that target the estrogen receptor).

You might say, it's a short treatment without any new fancy drugs. What's important about this trial? 2/
The key thing about this trial is that it is DESIGNED and POWERED to answer a question specifically for #lobular. Similar trials have been done, both the short- and long-term settings, but trials all had few (if any) ILC, so no strong conclusions for #lobular could be made. 3/
