Excited to share our new paper out at @NatImmunol today! We discovered the limiting factor for #brain #Tregs is #IL2, and developed a new #genetherapy to treat #traumaticbraininjury and other #neuroinflammatory pathologies. 1/12

The work started more than 10 years ago, after my brother Russell died following a #traumaticbraininjury. The more I read into it, the more treatable #neuroinflammation seems. Obviously there is a #brain delivery problem, but stopping #inflammation is basic #immunology! 2/12 Image
We had a great post-doc, @EmanuelaPasciu1, drive a project showing #Tcells in mouse and human brain, with key functions. Among these T cells were a small population of anti-inflammatory #Tregs, again in mouse and human. 3/12

Study from #England looking at the impact of #SARSCoV2 on #brain #pathology in 785 UK Biobank participants (aged 51–81) imaged twice found:

-brain changes in the following regions:
--greater reduction in grey matter thickness and tissue-contrast in the orbitofrontal cortex & parahippocampal gyrus
--greater changes in markers of tissue damage in regions functionally-connected to the primary olfactory cortex
--greater reduction in global brain size.
- infected participants also showed on average larger #cognitive decline between the 2 timepoints
-these imaging and cognitive longitudinal effects were still seen after excluding the 15 cases who had been hospitalized.

Happy 2021! @MBVanElzakker and I are excited to share our new article published in #Immunometabolism: “Pathogens Hijack Host Cell Metabolism: Intracellular Infection as a Driver of the Warburg Effect in Cancer and Other Chronic Inflammatory Conditions”: ij.hapres.com/htmls/IJ_1341_…
2/ In the paper, we detail molecular mechanisms by which #viral, #bacterial, and #parasite intracellular pathogens can induce, or contribute to, a Warburg-like #metabolism in infected host cells in order to meet their own replication and nutritional needs.
3/ We also discuss how host defense towards #infection may impact cellular metabolic changes (including how #mitochondria can participate in the innate immune response towards infection)

Ooof interesting! This team found Ljungan #virus (LV) antigens in autopsied #brain tissue from the hippocampus region of 18 #Alzheimer’s brains. No LV antigen was identified in any of their 11 age-matched control brains: ncbi.nlm.nih.gov/pmc/articles/P…
2/ The LV virus antibodies reacted with #neurons and #astrocytes and also showed distinct positive reaction in the #amyloid/neuritic plaques. The team thinks the findings indicate LV #virus may persist in the Alzheimer’s brain
3/ Here’s an image from one of the #Alzheimer’s brains. The red stain visualizes presence of LV viral antigen in neurons, astrocytes + #glial cells. In Panel C, an amyloid/neuritic plaque is shown staining positive in the glial compartment + in dystrophic neurites Image

Interesting! 👉 This study in mice found that persistent herpes #virus (HSV) activity detected in the brainstem...may primarily result from the transport of reactivated HSV from the trigeminal nerve into the brainstem: jvi.asm.org/content/jvi/ea… ImageImageImageImage
Context on the study 👉”Investigating HSV reactivation in the #brainstem is a reasonable first step to understanding #HSV pathology in the CNS, since...lesions in the front of the brain, characteristic of Alzheimer’s, may originate from #neurons located in the brainstem.”
But as the team explains, the study of HSV’s activity + reactivation in the central nervous system can inform the study of many #neuroinflammatory conditions (in addition to Alzheimer’s)
