Several studies have revealed that zinc does indeed improve semen volume and testosterone, and have explained the science behind how it works.

It had been known since the 1920s that zinc was essential for proper reproductive function in animals.

Zinc was especially concentrated in the testes, and would rise during mating season.

The first study in humans (1977) showed that 100 mg of zinc daily could massively improve testosterone:

400 ng/dL ➜ 815 ng/dL in zinc group
547.5 ng/dL ➜ 320 ng/dL in control

It also lowered pituitary gonadotropins.

Later studies showed that zinc could not only boost T, but improve other measures of male sexual health.

Low T men given ~44 mg zinc for under 2 months:

‣ More than DOUBLED sperm count
‣ Increased testosterone by over 50%
‣ Increased DHT by over 30%

9/22 men got their wives pregnant, when none could for 5 years prior.

Other trials confirmed this.

In another study, 50 mg as zinc acetate:

➞ Raised T
➞ Doubled sperm count
➞ Lowered gonadotropins (LH, FSH)

Direct and tight correlation between plasma zinc levels and testosterone.

Sperm quality is highly dependent on zinc.

Men with good sperm motility have much higher zinc levels in the semen.

WHY zinc is so good for male sexual health:

The main and most obvious role zinc plays is in antioxidant defense.

It is a cofactor for the enzymes superoxide dismutase and metallothionein, which prevent excess reactive oxygen species from damaging cellular compartments in the testes, and driving inflammation in them.

This oxidative stress will directly kill sperm cells, and suppress the expression of steroid producing enzymes.

Zinc also plugs into various enzymes and genetic regulatory proteins that support sexual health.

It supports the entire hypothalamic-pituitary-gonadal axis:

➜ Stimulates GnRH release and supports LH/FSH secretion
➜ Enhances Leydig cell sensitivity to LH, allowing more testosterone output with less stimulation
➜ Boosts transcription of steroidogenic genes like StAR and CYP11A1
➜ Inhibits aromatase, preserving testosterone from conversion to estradiol
➜ Maintains androgen receptor structure and DNA-binding

Zinc also protects the testes directly:

➜ Preserves Sertoli cell (sperm producing cells) function and the blood-testis barrier
➜ Concentrates in seminal fluid, supporting sperm motility, morphology, and DNA integrity
➜ Prevents apoptosis of developing germ cells
➜ Suppresses inflammatory cytokines like IL-1β and TNF-α that impair steroid production

Zinc deficiency leads to high LH/FSH but low testosterone, indicating resistance in the axis. Zinc repletion restores sensitivity, boosts testosterone, and revives spermatogenesis.

A recent study showed that rinsing with coconut oil was superior to conventional mouthwash for dental health (periodontitis).

The study was published in March in the Journal of Clinical Oral Investigations.

People with periodontitis were given either:

➞ Standard treatment (chlorhexidine mouthwash)
➞ Coconut oil mouthwash (5 mL, 10 mins / day)
➞ Placebo mouthwash

For a month. They were checked in a month after treatment stopped.

Coconut oil maintained diversity of the oral microbiome.

In both saliva and gingival crevicular fluid, indices of bacterial diversity declined in both placebo and in standard treatment, but not with coconut oil.

This is considered good - as coconut oil did not wipe out bacteria considered beneficial.

It examined giving:

◇ Glycine: an amino acid in foods like collagen

&

◇ N-Acetylcysteine: a modified form of the dietary amino acid cysteine

To older adults.

Both at a dose of 100 mg / kg bodyweight, daily, pretty big doses of both.

Older adults were randomly put in either the glycine + NAC (GlyNAC), or a placebo (alanine).

The study was 12 weeks.

They also had younger adults enroll to see if the supplementation could really make them biologically resemble youth.

A fascinating study showed that Vitamin E nearly doubles testosterone in response to stimulation, and by over 100 points at baseline.

Subjects were restricted to eating the diet the hospital put them on, with a consistent amount of vitamin (3-5 mg).

They were then put on 483 mg (724.5 IU) of vitamin E for 8 weeks.

This is a huge dose - over 30X the "recommended" daily amount.

It was also vitamin E acetate - a relatively poorly absorbed form of the vitamin.

After just 2 weeks - the high dose vitamin E was shown to increase both total and free testosterone substantially.

Total T went form 416 to 545 by the end of the 8 weeks.

The free T index increased by ~28%.

No changes in LH - indicating that the sensitivity of the testosterone producing Leydig cells was improved.

A number of eye opening studies show that biotin (vitamin B7) megadoses can dramatically improve diabetes, and even reverse it.

The most striking of the bunch was done in 1993.

People with type 2 diabetes (>171 mg/dL fasting blood sugar) were recruited.

These people were on sulfonylurea drugs or dietary treatments, with little to no success in controlling their diabetes.

People either took 9 mg (a HUGE dose) of biotin or placebo.

People with diabetes had less than 60% of the serum biotin that healthy people did.

This was strongly correlated to higher blood sugar.

The higher the biotin level, the lower the blood sugar.

The study was published in The Journal of Trauma, Injury, Infection and Critical Care.

While it was a relatively small pilot study, the results were so encouraging that it's a shame it's never been properly followed up on, as the study was published in 2006.

They took children and adolescents who had severe traumatic brain injury.

Within 4 hours of the injury, they received 0.4 g / kg bodyweight of creatine, for 6 months.

This is a pretty huge dose, 4-5X the amount that is typically recommended for athletic performance.
