DBPs in Shilajit (and why they're so interesting)

Most people talk about how Shilajit contains a lot of trace minerals and Fulvic acid.

But the most interesting compounds that can be found in Shilajit are the DBPs, which are also known as:
Dibenzo-alpha-pyrones

Why are these DBPs so beneficial?

there are 3 big reasons

(boring science warning)

NUMBER ONE:
they're 'Quinone-like' compounds

DBPs are technically not Quinones, because they do not have the structure of a Quinone (their core structure is a fused tricyclic lactone and not a cyclic dione)

But they act like quinones in redox reactions, electron transport, and mitochondrial function due to how they cycle between three redox states (reduced, semiquinone radical, and oxidized/quinone-like forms).

Accepting electrons from Complex I and II →  then carrying those electrons to Complex III → in Complex III the electrons are donated and carried to Complex IV where the electrons are handed off to Oxygen preventing electrons to accumulate at Complex III and excess ROS to be produced

NUMBER TWO:
They work in tandem with CoQ10

CoQ10 shuttles electrons between Complex I/II & III but can be easily oxidized back from Ubiquinol (active CoQ10) back into Ubiquinon (oxidized form) under stress.

DBPs prevent the oxidation of Ubiquinol into Ubiquinon, and keep CoQ10 in its reduced form, protecting it from oxidative stress.

What does this mean?
Ubiquinol stays active for a longer time duration in the ETC (electron transport chain) causing less electron leakage and increased ATP production

Ubiquinol is kept in the reduced form and its ability to neutralize free radicals is extended because of how Ubiquinol acts as antioxidant

Mitochondrial membrane potential is now better preserved

Energy production stays stable and efficient under stress, heavy exercise or even prolonged period of focus and attention

Can handle more exercise, more work and even more emotional stress

NUMBER THREE:
Mitophagy benefits of DBPs

Mitophagy is the process where your cells detect/recycle damaged/dysfunctional mitochondria through autophagy which prevents excessive ROS/oxidative stress and ensures that inefficient mitochondria are cleared away, keeping energy production efficient.

This helps to prevent fast aging, low energy and diseases.

So how does Shialjit enhance mitophagy through DBPs?

One of the primary DBPs in Shilajit is:
‘3,8-dihydroxy-6H-dibenzo[b,d]pyran-6-one’

this DBP is chemically similar to Urolithin A

It stabilizes PINK1 on damaged mitochondria, which then helps to recruit ‘Parkin” which then makes the mitochondria a visible target for mitophagy by attaching ubiquitin chains to the many proteins that sit on the outer membrane of the mitochondria.Image

Intranasal Insulin seems to be something very niche, until you realize that there's a tremendous amount of scientific literature on it, just go on google scholar and type in 'intranasal insulin' and you'll see.

Seems to be mainly mediated through improving redox balance in the brain, just like Cerebrolysin

no, intranasal insulin does not cause any insulin resistance (as far as we know)

Bee venom against fibrosis/scar tissue formation in the scalp:

Male pattern baldness is a slow process of scar tissue formation in the scalp (fibrosis), eventually when the amount of scar tissue starts accumulating more and more, it starts to suffocate the hair follicles, and they die off.

This entire process is mediated by a ton of hormonal inputs, inflammatory cytokines and hypoxic markers, but they all pretty much exert their effect on speeding up male pattern baldness by upregulating TGF-beta1.

TGF-beta1 is the master regulator of Fibrosis (scar tissue formation), its activation leads to:
→ fibroblast activation
→ ECM deposition
→ myofibroblast differentiation

Reducing expression of TGF-beta1 is key when it comes to putting a halt to male pattern baldness.

And Bee stings reduce TGF-beta1 expression exceptionally well through Melittin by inhibiting NF-κB activation (through blocking IκB degradation) and suppressing the consequent release of various pro-inflammatory cytokines like TNF-alpha, IL-6 and IL-beta1 as well as through the direct interference of Melittin with the TGF-beta1 receptor itself.

Bee venom promotes blood flow in scalp:

Bee venom locally upregulates VEGF, a growth factor which dictates the process of angiogenesis, this is the process where new capillaries (very small blood vessels) are formed locally.

VEGF is already produced by cells in the hair follicle during the Anangen (growth phase) to enhance vascularization which helps with improving blood flow to the hair follicles that delivers the necessary oxygen and nutrients to grow healthy (and stay alive).

VEGF its expression is higher when the hair follicle is in the Anagen (growth) phase, starts declining in the Catagen (regression) phase, and eventually becomes very low during the Telogen (rest) phase.

Bee venom its ability to potently enhance local VEGF expression ultimately helps with improving vasculature of the scalp and allows for hair follicles to get the necessary nutrients needed for growth.

There’s nothing with a more neurotoxic profile and a dimming effect on the soul like viewing yourself as a loser has.

viewing yourself as a loser actually has a scientific term → ‘social defeat stress’

you essentially view yourself as a victim to your external environment (lack of control equates to learned helplessness) and at the bottom of the social hierarchy.

This feeling of not being in control of situations, or lacking a strong future vision, weakens activity in the prefrontal lobes (mainly mPFC, vmPFC and dlPFC).

These prefrontal cortexes normally have an inhibitory effect on the Amygdala, but when this inhibition fades due to negative self-talk and surrender to external environment, it causes the gut-microbiome its alpha-diversity to decrease through HPA axis activation.

result?

reduced SCFA production, with Butyrate in particular.

Butyrate is crucial for:

- HDAC inhibition (intelligence, memory, focus, motivation)
- androgen receptor expression
- reducing estrogen receptor expression through hyperacetylation
- increasing natural Testosterone production through the HPG
- thyroid hormone uptake

Back in 2022, a bunch of researchers showcased how they were able to completely alter the social dynamic between two mice by simply altering their gut-microbiome through antibiotics, targeting C. Butyricum in particular (one of the main bacteria that produces Butyrate)

The researchers could turn an ‘alpha’ mouse into a ‘beta’ mouse, making it subordinate to the mouse that was previously ‘beta’ in the social dynamic.

It was found that recolonization of the gut-microbiome of said mouse that ‘turned beta’ with C. Butyricum could restore that mouse’s status, and make him ‘alpha’ again.

The researchers thought this was because of the huge role that SCFA’s, with butyrate in particular, play in activity of the prefrontal cortex (mPFC, vmPFC and dlPFC) due to Butyrate its function as HDACi.

With insufficient SCFA/Butyrate levels, it’s almost guaranteed that the function of the prefrontal cortex is going to be impaired in some way.

Results:

- less discipline
- lack of willpower
- reduced goal-achieving capabilities
- surrender to reptile brain (prefrontal cortex exerts inhibitory control over these)
- ADHD-like behavior
- excess rumination of thoughts/overthinking (negative spiral)

Height-growth DOESN'T have to stop AFTER the growth plates have closed, and the science points towards that.

Let's start by looking at the fingers.

Which still shows increases in length even after the growth plates have fully fused

This study found that after the age of 21 (age where the growth plates have fused) the length of the finger continues to increase in size with the normal aging process.

This increase in finger size was much higher in males that worked manual labour, which is a direct argument for Wolff’s law and how mechanical stress influences bones, even at later ages.

This increase in finger length is tiny, but the fact that the length keeps increasing, even when the growth plates have closed down, goes to show that bone doesn’t just lengthen only from the growth plates.Image
The size of the fingers/hands KEEPS increasing AFTER all the growth plates in the hands have fully fused, and the hands/fingers actually keep growing in size even 4 YEARS after the growth plates have fully closed.

With some increases in size even going as high as 5 whole centimeters (2 inches for my american friends), that’s not miniscule, it’s actually quite significant, and the bone growth happens AFTER the plates were closed.

How would this be possible when the growth plates were closed?

In a 2009 study conducted in Serbia on 12 year old boys, researchers found Royal jelly to be one of the strongest foods to enhance growth when still in puberty.

This is how the study went:

the research was carried out with 25 football players (n=25) from FC "Red Star" Belgrade which all were 12 years old.

The football players were seperated into two groups:
- control group that didn't use Royal Jelly (10 people)
- experimental group that DID use Royal Jelly (15 people)

All 25 players from both the control and experimental group were tracked during a 2 month period where they trained 4 times a week and competed in the youngest Belgrade league.

The results were astonishing.

The average height of the control group went from 158,12 centimeters to 158,82 centimeters, a total increase of around 0,7 centimeters.

The average height of the research group went from 152,46 to 154,13 centimeters, which is a staggering increase of 1,6 centimeters in two months time.

But not only did the research group (the ones that took Royal Jelly) gain a more profound increase in height.
