Fantastically optimized and detailed single cell-RNA-seq protocol (variant of sci-RNA-seq3) developed by @bethkarenmartin, out in @NatureProtocols today, less than 1 cent per cell.
tinyurl.com/34a9e8tp This protocol was used for both in vivo and synthetic tiny mouse embryos in studies from @CXchengxiangQIU and @ZernickaGoetz.

Including stuff in the works Beth alone has profiled >10 million nuclei w/ this protocol.

Short thread on our latest work, from the incredible @CXchengxiangQIU (CX Qiu), entitled "Systematic reconstruction of cellular trajectories across mouse embryogenesis", which is out today in @NatureGenet. Link to accessible version: 1/n
rdcu.be/cIWq2 Backstory: In 2019, with @junyue_cao @malte_spielmann @coletrapnell, we profiled 2M cells from mouse E9.5-13.5. Incidentally, @MarioniLab @reik_wolf & others profiled earlier windows. Collectively these spanned E3.5-13.5 (10 days in which embryo goes from 32 to >10M cells!) 2/n

Today we release in @biorxivpreprint a pair of related methods, DNA Ticker Tape & ENGRAM, for multiplex, temporally ordered genomic recording of cell lineage histories, enhancer & signal transduction activities, respectively led by the brilliant @_Choi_Junhong & @wchenomics 1/25 Links to preprints on @biorxivpreprint:

ENGRAM: tinyurl.com/2784mhkh

Let’s start w/ DNA Ticker Tape, by @_Choi_Junhong, a general system for pseudo-processive genome editing that can "write" the ORDER of molecular events. 2/25
