Zhou Yusen's vaccine construct (CN111333704B) — present in December 2019 BALF, two months before the patent filing.

111 genome assemblies: near-complete nucleotide clonality across RBD and nsp12. Inconsistent with four months of natural circulation.

Finding 5 of 7. The molecular cloning signatures.

SARS-CoV-2 has four spike insertions relative to RaTG13, its closest known bat coronavirus relative. Two of them — unique to SARS-CoV-2, absent from RaTG13 — carry restriction enzyme recognition sequences at their boundaries.

Flanked by NcoI-compatible sequences (CCATG / CATGGT).

NcoI recognition site: CCATGG. Standard cloning enzyme for expression vector insertion. The NcoI site generates a 4-nt CATG overhang used for directional cloning into hundreds of standard laboratory vectors.

This insert does not exist in RaTG13. The NcoI site does not exist in RaTG13.

That's the line between library prep contamination and a construct inside the patient.
