The best way to find out this significance of King's tattoo is to look at what the reference might be and what it might represent.
Many have already noticed, from when we got the first glimpse of it, that it looks similar to a Laurel Wreathe.
This has already been pointed out by people like, @Peronas_Ghost, who theorised that King may have an association with Emperor Nero.
This seems like it is a good idea and I think even if it is not exactly a representation of a Laurel Wreathe, the idea that it is referencing Old Testament mythos seems to be leading us along the right lines.
This line of thinking is backed up by the recent revelation of King's real name, Arber / Arbel. Depending on the proper pronunciation, this may mean different things.
If the proper pronunciation is, 'Arbel', that is an Israeli name meaning, 'Divine Night', this would fit with the idea that King is a Lunarian and that the Lunarians were once regarded as Gods.
However, if it is prononced, 'Arber', then that means 'tree' in German, from the Latin root word, 'arbor'.
If this is the case, then it may give us the connection to Shandora and the reason why these places are known as 'Holy Lands'.
This was actually explained in Skypiea.
During the celebration before the expedition to find Shandora, Gan Fall explained to Zoro and Robin, why Jaya was also known as, 'The Holy Land.' Because it's rich soil allowed them to grow plants, something that was not possible in the sky.
They called this, 'Vearth'.
This information is given further context through Enel's cover story, through which we learned that the 3 winged races came from the moon city of Birka, seeking natural resources.
With this in mind, it becomes clear why the Lunarians settled on the Red Line.
As Nami explained, the Red Line offers all climates, this would make it an ideal place for the Lunarians. And the peaks of the Red Line would be at the perfect altitude for a winged race like the Lunarians.
It seems likely that the Lunarians prospered through plants.
This then, explains that Jaya and Mariejois, are known as 'Holy Lands' for the same reason, these lands allowed the winged races to grow their natural resources.
It seems likely that this was the pride of the Lunarian civilisation, as it was for the Shandorians and Skypieans
It seems no coincidence then, that within Mariejois, there is a place called, 'The Room of Flowers.'
This was likely the Lunarian Garden.
Many have already theorised that this room is a reference to, The Garden of Eden.
If this is the case, then perhaps that provides the connection to the tattoos of Calgara and Wyper, they may represent the Devil Snake that tempted Eve with the forbidden fruit.
It seems likely that within this garden, there is a particular tree. Many have already suggested, that there may be a Devil Fruit tree in the Room of Flowers. But perhaps there is a tree that references, The Tree of Life, that appears in Genesis.
This connection may provide us with a huge clue as to what King's Tattoo may represent, the blossom of a particular tree, a 'Tree of Life.'
A Cherry Blossom.
It's easy to forget that Dr Hiriluk told the story of a famous pirate from West Blue, who suffered an incurable heart disease.
It was only thanks to miracle that he survived. He stumbled across a forest of Cherry Blossoms.
Shortly after, he was cured.
It is for this reason that Hiriluk chose the Cherry Blossom as the symbol of medicine.
