I designed intentional "Twitter Time Breaks" throughout my presentation for audience members to engage, respond to questions, share ideas, interact with each other on Twitter, etc. I told the audience about this right at the start of the presentation.
Rather than designing a unique hashtag to facilitate this, I decided to keep all interaction within one tweet posted on my Twitter account. Audience members could "reply" throughout the presentation to this tweet rather than having to post their own tweets from their profile.
