Sotrovimab, a COVID-19 antibody treatment that has been approved in the UK, has proven to be effective against ALL new mutations in the spike protein of the Omicron variant, NOT just the key mutations. The tests included ALL 37 mutations identified to-date in the spike protein.
Sotrovimab reduced the risk of hospitalization and death in people with mild to moderate COVID-19 by 79% in trials.
Given the less than three-fold drop in neutralization during tests, developers are confident that Sotrovimab will continue to provide significant benefit for the early treatment of patients hoping to avoid the most severe consequences of COVID-19. The injected antibodies are just
one tool in the arsenal of COVID treatments, but could be key to immune-suppressed people who don’t mount an adequate response to vaccines and help lessen the effects of the virus in those more susceptible to severe disease.

Interesting fact. Sotrovimab was cloned from memory B-cells of an individual who recovered from COVID-19, many recipients of our current vaccines will develop similar antibodies. Immunological memory for the win! Past studies: nejm.org/doi/full/10.10… and nejm.org/doi/full/10.10…
ADDITION: Sotrovimab is authorized for use under an Emergency Use Authorization (EUA) for the treatment of mild-to-moderate COVID-19 in adults and pediatric patients with positive results of direct SARS-CoV-2 viral testing, and who are at high risk for progression to severe

Anyone who is interested in a well-written, informative thread regarding some positive signs on Omicron and severity read this.
Some takeaways:
•Share of COVID-positive patients in ICU or on ventilators is somewhere between 2-3x lower than it was at the same stage of the Delta wave
•Suggests reduction in severity of outcomes, and we see something similar when we look at length of stay in hospital data
•Data on admissions and discharges shows there is a clear gap between the two and is shorter for this wave, suggesting milder illness
•Unlike total COVID-positive patient numbers, which are close to Delta levels, the numbers in ICU and on ventilators are low
•Majority of patients in COVID ward were actually admitted for other reasons (all receiving oxygen for COVID were unvaccinated)
•Cautious optimism is key
•Omicron being “milder” seems to be consistent with immune protection so, with that said, make sure you’re protected!

World Health Organization: Preliminary data indicates hospitalizations in South Africa remain LOW, and emerging data indicates Omicron may cause LESS severe illness and is MILDER, but possibly more transmissible. Immunity through vaccination and previous infection is kicking in!

Fantastic news. Initial lab studies show a third dose of Pfizer’s COVID-19 vaccine neutralizes Omicron. A booster with the current version of the vaccine RAISES antibodies 25-FOLD providing a similar level as observed after two doses against the original virus and other variants!
As 80% of epitopes in the spike protein recognized by CD8+ T-cells are not affected by the mutations in Omicron, two doses MAY STILL induce protection against severe disease.
Blood plasma from people immunized with two doses of the vaccine had a 25-fold reduction in neutralizing antibody levels versus Omicron, when compared with the original strain of the virus, boosters RESTORE that loss.

Preliminary data on Omicron and whether it escapes vaccine antibodies shows immune escape is “robust” but NOT complete- it’s partial! Reduction in nAbs was even LESS in those who had a previous infection & two doses of Pfizer’s vaccine. Boosters should be able to take this on!🧵
VERY prelimary data so numbers subject to change but live virus studies show a 20-40 fold reduction in nAbs with two doses of Pfizer’s vaccine. ONLY TWO. This is NOT regarding boosters. CRUCIAL point: the reduction was not total. It’s partial. I know the number sounds alarming-
BUT that DOES NOT mean 40%-60% decrease in vaccine efficacy! Please don’t get confused. I told you guys these vaccines could take a hit and still hold up. Now, let’s continue.

Now, if one is wondering why hospitalizations are going up, according to their MOH, 90% of hospitalizations in South Africa are unvaccinated individuals (zero doses), 10% remaining are mostly partially vaccinated (one dose) Low vaccination rates (less than 30% fully vaccinated).
In regards to the rise in cases, this isn’t exactly surprising based on the study I highlighted yesterday on Omicron’s ability to reinfect. Per in article in this thread, hospitalizations seem to be lower proportion of cases than previous waves. medrxiv.org/content/10.110…
