FANTASTIC news for your Friday! Phase 2/3 results of PAXLOVID, Pfizer’s COVID-19 oral antiviral shows the risk of hospitalization or death is REDUCED by NEARLY 90% in high-risk adults with COVID-19 when taken within three days of experiencing symptoms.

Let’s talk about that! 🧵
IMPORTANT NOTE: this protease inhibitor was originally developed for SARS-CoV in 2003 and has been repurposed! So first, let’s discuss the results. The scheduled interim analysis showed an 89% reduction in risk of COVID-19-related hospitalization or death from any cause compared
to placebo in patients treated within three days of symptom onset (primary endpoint); 0.8% of patients who received PAXLOVID™ were hospitalized through Day 28 following randomization (3/389 hospitalized with NO deaths), compared to 7.0% of patients who received placebo and
were hospitalized or died (27/385 hospitalized with 7 subsequent deaths). In the overall study population through Day 28, NO deaths were reported in patients who received PAXLOVID as compared to 10 deaths in patients who received placebo.
PAXLOVID is an investigational SARS-CoV-2 protease inhibitor antiviral therapy, specifically designed to be administered orally so that it can be prescribed at the first sign of infection or at first awareness of an exposure, potentially helping patients avoid severe illness
which can lead to hospitalization and death. How does it work exactly? It is designed to block the activity of the SARS-CoV-2-3CL protease, an enzyme that the coronavirus needs to replicate. Co-administration with a low dose of Ritonavir helps slow the metabolism of it to
remain active in the body for longer periods of time at higher concentrations to help combat the virus. PF-07321332 inhibits viral replication at a stage known as proteolysis, which occurs before viral RNA replication. In preclinical studies, PF-07321332 DID NOT demonstrate
evidence of mutagenic DNA interactions. At the recommendation of an independent Data Monitoring Committee and in consultation with the U.S. Food and Drug Administration (FDA), Pfizer will cease further enrollment into the study due to the overwhelming efficacy demonstrated in
these results and plans to submit the data as part of its ongoing rolling submission to the U.S. FDA for Emergency Use Authorization (EUA) as soon as possible. If approved or authorized, PAXLOVID, which originated in Pfizer’s laboratories, would be the first oral antiviral of
its kind, a specifically designed SARS-CoV-2-3CL protease inhibitor. Upon successful completion of the remainder of the EPIC clinical development program and subject to approval or authorization, it could be prescribed more broadly as an at-home treatment to help reduce illness
severity, hospitalizations, and deaths, as well as reduce the probability of infection following exposure, among adults. It has demonstrated potent antiviral in vitro activity against circulating variants of concern, as well as other known coronaviruses, suggesting its potential
as a therapeutic for multiple types of coronavirus infections. It’s now the second antiviral pill behind Merck’s to demonstrate strong effectiveness for treating Covid at the first sign of illness which was just approved by the MHRA: gov.uk/government/new….
So how does this all work? REMEMBER, this is NOT meant to REPLACE a vaccine. Vaccination is still our best tool to reduce the risk of infection, hospitalization, and death due to COVID-19. They’re PREVENTATIVES. IF infected, whether due to being unvaccinated OR a “breakthrough”
infection, and you are experiencing symptoms these antivirals can help immensely to reduce the risk of hospitalization EVEN further. With that said, here is to a BRIGHTER 2022! See: washingtonpost.com/health/new-cov… and pfizer.com/news/press-rel…. EDIT: Merck’s was repurposed not Pfizer’s.

Just a friendly reminder. Despite what you might hear, there has yet to be a variant that isn’t susceptible to the vaccines.
Why are variants unlikely to FULLY evade vaccine-induced immunity?
•Vaccines are polyclonal
•CD8+ T-cells covering 52 epitopes across the spike protein
•CD4+ T-cells covering 23 epitopes across the spike protein
You can learn about epitopes here: news-medical.net/life-sciences/…
Regarding the concerns about waning immunity. Please remember, this is likely referring to infection. Not effectiveness against symptomatic infection, not effectiveness against severe illness. See: bmj.com/content/374/bm…

“The vaccines don’t work- look at Singapore.” Alright, let’s.

Cases are at record highs due to Delta, however, ALMOST 99% of Singapore’s vaccinated cases are MILD or have NO symptoms. In addition, the nation has had VERY FEW deaths despite a surge in cases.

I’m not done.🧵
Almost 84% of Singapore’s population is considered to be fully vaccinated. IMPORTANT TO NOTE: Singapore transitioned away from a ZERO COVID approach. The rise in cases upon attempting to reopen was to be expected as part of attempting to “live” with the virus and reopen.
According to their MOH, over the last 28 days, of the 96,376 infected individuals, 98.7% are MILD or have NO symptoms, 0.8% required oxygen supplementation, 0.3% required ICU care, and 0.2% had died. moh.gov.sg/news-highlight…

Let me fix this BBC headline for you since most unfortunately will not read past it, OR even read the study. Fully vaccinated individuals can contract and pass on COVID-19 in the home, BUT AT LOWER RATES than unvaccinated individuals. bbc.com/news/health-59…

I’m not finished.🧵
If you actually open this article, rather than just posting a screenshot of it and taking it as some sort of sarcastic “win” as I have seen several people do this morning, you will find that this article does not really say what you think it says and I think most people
are counting on the fact most will not read past the headline. So, what does it actually say? First and foremost, and from the author of the study themselves: “The ongoing transmission we are seeing makes it ESSENTIAL for unvaccinated people to get vaccinated to

A study of 1M+ people in Canada found two doses of any of the three COVID-19 vaccines (Moderna, Pfizer, AstraZeneca) led to strong protection from infection, hospitalization, AND death -even against Delta- REGARDLESS of the vaccination combination and showed NO signs of waning.🧵
Data from researchers at the British Columbia Centre for Disease Control (BCCDC) and the Quebec National Institute of Public Health (INSPQ) showed two doses of mRNA and/or AstraZeneca’s vaccine gave excellent protection against hospitalization, with NO SIGN of decline
by 5-7 months post-vaccination. In addition, a 7-8-week interval between doses improved vaccine efficacy and may be optimal in most circumstances. Findings indicate prolonged two-dose protection and support the use of mixed schedules and longer intervals between doses.

In case it needs to be said, COVID-19 vaccines DO NOT and WILL NOT cause AIDS. This false claim has been circulating due to the manipulation of documents which DO NOT even mention anything of the sort in their original text. Stop the misinformation.
Please remember, the risk of developing aids comes through infection with HIV, which can occur from exposure to infected bodily fluids OR by sharing needles with someone who is infected.
At NO time will your immune system weaken due to the COVID-19 vaccine or any vaccine for that matter. Vaccines encourage and train our immune systems, not the opposite. In addition, please remember, neutralizing antibodies always decline, AS THEY ARE SUPPOSED TO! But guess what
