Many viruses enter the human host through mucosal surfaces. Local immunity at the site of virus entry provides superior protection than systemic immunity. Here is my take on mucosal immunology and how it can be leveraged for vaccine development.

There are many innate and adaptive effector mechanisms that can block virus entry, infection and spread. Vaccines that elicit mucosal IgA, IgG, tissue-resident memory T cells are ideal to combat mucosal viral infections. (3/n)
Immune correlates of protection for most vaccines are measured by the titers of antibodies they produce in the serum (even for mucosal viruses). Neutralizing antibody titers in the serum are a proxy for the levels of #NAb needed for protection at mucosal surfaces. (4/n)
Vaccines can elicit stronger immune response than natural infections. Here is a nice example from human papillomavirus (HPV) vaccine.

Thus, #COVID19 vaccines can (and should) induce more robust and durable protection than natural infection. (end)
