STOX1 is a TF and predicted #preeclampsia risk gene IDed through family-based genetic linkage studies (van Dijk, @NatureGenet 2005). A variant in the DNA-binding domain was proposed to alter protein function ➡️ preE susceptibility. But LOF model had not been studied. 🐁 2/
*We found that loss of Stox1 ➡️ gHTN assd w placental hypoxia + abnormal ECM deposition.
*We observed activation of the uteroplacental renin-angiotensin system in Stox1 KO mice.
*Treatment w an ARB, losartan, rescued the gHTN phenotype, validating the mechanism. 3/

Genetic mouse models are invaluable for studying mechanisms involved in hypertensive disorders of pregnancy.
The @preeclampsia spectrum is broad and still poorly understood. More basic science in this area, please!
