RT – FINALLY out! Our new genetic study of number of children born greatly extends our 2016 Nature Genetics GWAS. Newly identified associations indicate genetic loci under present-day natural selection - here is a summary thread....biorxiv.org/content/10.110…
Extending our 2016 Nature Genetics GWAS of number of children nature.com/articles/ng.36… with a sample of around 785K, we find 43 loci associated with number of children ever born or childlessness

We show that diverse biological mechanisms are related to reproductive success – both behavioural and neuro-endocrine influences biorxiv.org/content/10.110…
We joined forces this time with @mathiesoniain & compared our genome-wide significant loci associated with number of children ever born and childlessness with ancient DNA over timescales from ~2,000 to ~30,000 years before present

The strongest overlap was at chr11:61.5MB with posterior probability of 96% that lead variants for ancient selection on number of children ever born represent same underlying signal biorxiv.org/content/10.110…
Integration with data from ancient DNA selection scans identifies that the FADS1/2 gene locus that has been under selection for thousands of years still remains under selection today
biorxiv.org/content/10.110…
Number of children ever born increasing alleles increased in frequency over recent generations. The derived allele increased by 0.0088% per-year between the 1938 and 1969 birth cohorts – corresponding to a selection coefficient of 1.2% (95% CIs -0.9-3.2%)
Missense alleles in ARHGAP27 are associated with increased number of children born but reduce reproductive lifespan – suggesting a trade-off between reproductive ageing and intensity biorxiv.org/content/10.110…
And for those poised to send me a long mansplaining email or tweet - yes, I have thought about how it is even possible to study the genetic transmission of having children & childlessness nature.com/articles/ng.36… or this nature.com/articles/ejhg2…
