This is the first #proteomic study that uses high throughput pQTL in neurological relevant tissues to identify novel genetic loci regulating protein levels, which will improve our understanding of the biology of #Alzheimers, #Parkinsons, & other neurodegenerative diseases
We identified hundreds of novel pQTLs in CSF, plasma, & Brain linking novel genes, biomarkers and drug targets with #Alzheimers, #Parkinsons
In this extensive proteomic study in cerebrospinal fluid, we identified hundreds of genetic regions modifying disease-relevant protein levels, including GRN, CD33, YKL40, & #APOE.
Mendelian Randomization applied to multi-tissue proteomic data identified several novel biomarkers for #Alzheimer's risk, onset, & progression, and other neurodegenerative traits, including #Parkinsons, Frontotemporal dementia, #ALS & #stroke.
Our multi-tissue pQTL indicates that only 30-40% of the pQTLs are also eQTL, suggesting that leveraging pQTL data can resolve additional GWAS signals for complex traits.
In the last 20 years, GWAS have uncovered ~40 loci associated with #Alzheimers. Many of those loci are multi-gene, our study shows that pQTLs is a valuable tool for #functional genomics & fine-mapping causal associations in complex diseases. @Alzheimers_NIH @alzforum
