Hunt for a universal #SARSCoV2 vaccine that will work regardless of spike mutations—next holy grail.

Instead of targeting the spike protein, which is prone to mutations, researchers are assessing other parts that are known to be more stable. 🧵#COVID19
cepi.net/news_cepi/goin…
2) “For example, scientists at University of Nottingham in the UK are working alongside pharmaceutical company Scancell and Nottingham Trent University to test its COVID-19 vaccine candidate, SN14, which targets both the spike protein as well as the nucleocapsid, or N-protein.”
3) “By targeting this additional viral structure, which is far less likely to mutate, the vaccine if found safe and effective could work to protect people against COVID-19 irrespective of any mutations to the spike protein and, in theory, could work across other coronaviruses.”
4) What can the ‘shape-shifting’ flu teach us?

Although the COVID-19 pandemic has sped up ideas to create a universal coronavirus vaccine, the drive to develop an all-in-one virus protection device isn’t new.
5) “The flu vaccine, for example, is designed to elicit antibodies capable of neutralising the virus if an individual becomes infected. However, given the ‘shape-shifting’ abilities of the flu virus—with its frequent mutations and hundreds of strains—
6) “the vaccine is delivered to populations on a seasonal basis, typically only tackling the predominant strain circulating that year and becoming ineffective in subsequent years against other strains.
7) “costs and difficulties of year-to-year approach (vaccines continually amended and immunisation campaigns annually), scientists have theorised that long-lasting immunity could be provided through a more broadly protective or even universal flu vaccine of different strains.”
8) “It would not only aid the responses to seasonal influenza but could also be critical tools for preventing a potential flu pandemic.”

And indeed— we are getting closer to a universal flu vaccine—see study below.
9) back to #SARSCoV2–“CalTech engineered a prototype all-in-one vaccine via a nanoparticle to hold fragments of the spike from multiple known coronaviruses—innovative platform was found to trigger the production of antibodies against several coronaviruses in preclinical testing.”
10) “Importantly, it not only protected against coronavirus antigens in the vaccine but also against related strains, suggesting an immune response had recognised common features across coronaviruses. This could therefore provide protection against newly emerging coronaviruses.”
11) “Another approach, suggested in a recent Nature commentary, could be to look at whether broadly neutralising antibodies, known to work against multiple HIV strains, also exist for coronaviruses. While neutralising antibodies typically defend the body from one antigen...”
12) “broadly neutralising antibodies can react to multiple antigens so could be used as the basis to design vaccines against a number of coronaviruses... people who have antibodies following earlier SARS infection also showing some protection against SARS-CoV-2.”

Increased mortality for #B117—New study in 🇬🇧 finds 64% higher risk of mortality for #B117 versus previously circulating variants (95% confidence interval 1.32–2.04) in ~55,000 people who tested #COVID19 positive in community. Risks grew after >12 days. 🧵

2) In this population, “this represents an increase in deaths from 2.5 to 4.1 per 1000 detected cases.”
3) “The matched cohort design controls for most potential biases, including variations in hospital capacity, as it pairs patients by personal characteristics, geography, and time of testing.” It also adjusted for age.

Engineer Allan J. McDonald, who in 1986 tried to stop the launch of the Challenger space shuttle, citing the effect of the cold on booster rockets, and who, after it broke apart on liftoff, blew the whistle when officials tried to cover, has died at 83. 🙏 nytimes.com/2021/03/09/us/…
2) “During the hearing, Ms. Ride asked Lawrence B. Mulloy, who oversaw the booster rockets for NASA, about rumors of a dissent by Morton Thiokol engineers. Mr. Mulloy conceded that there had been a discussion, but said that the company had ultimately agreed to go ahead....
3) “At that point Mr. McDonald, sitting in the back of the room, stood up. His hands shaking, he told the panel that Mr. Mulloy was not giving them the whole story; the engineers, he said, had been pressured and overruled.
Mr. Rogers immediately asked for the room to be emptied

💡AWESOME FOR BABIES—Experts & new studies suggest pregnant women who recovered from #COVID19 or vaccinated are likely to pass on protective antibodies to fetuses in their womb & to babies via breastfeeding—protects babies both during & after pregnancy.🧵

2) “Researchers in Oregon recently analyzed milk samples from a group of six mothers, half of whom received the Moderna vaccine and the other half who received the Pfizer version.

The milk tested negative for antibodies before the mothers were vaccinated, and spiked afterward.”
3) “Breast milk samples were collected pre-vaccination and at 11 additional time-points, with the last sample taken 14 days after the second vaccine dose. No adverse effects were discovered among the babies.”

The pandemic is devastating the world economy—however, good news is US’s Biden stimulus of $1.9 trillion will not only help save US but also help the world. And vaccine rollout is critical.

But how much depends on how fast we all #COVID19 vaccinate.

2) but one major country will likely re-emerge by the end of 2021 back to 2019 GDP growth levels... because of the Biden’s $1.9 trillion stimulus... and if we keep up the pace of vaccinations.
3) Here is the huge impact of the Biden COVID stimulus. The worry is that Euro zone and Japan might fall behind.

⚠️HUGE INCREASE IN KIDS—New Swedish 🇸🇪 data showing a sudden dramatic surge in pediatric #COVID19 cases across Sweden since Jan.

Kids age 0-9: Up ⬆️ 123%
Kids age 10-19: Up ⬆️ 72%

Likely due to surging #B117 variant, where it’s now 50% in Stockholm.🧵

(ht graph @DavidSteadson)
2) Is this pediatric surge in #COVID19 a statistical fluke? No. If we just look at the change from late Feb to first week of March, we still see this consistent surge in kids.

Also, @DavidSteadson notes, while many schools were open, high school grades 10-12 closed in person.
3) And where is the proof that #B117 is surging fast in Sweden 🇸🇪? Here on Swedish Public Health Authority’s website. Circled is Stockholm’s 53% #B117 proportion found in week 8 of 2021. Another region has 60%.

This #B117 surge is a real concern.
