In the #nanoporeconf data theatre with @Scalene talking about our recent Gigascience paper on sequencing the Zymo mock community, and his new DNA extraction method academic.oup.com/gigascience/ar…
Introducing the "three peaks challenge", three DNA extractions to ensure representation. "it's not ok to sacrifice representation to get long reads"
The name is a bit of an in-joke as we only really see two peaks in the read length distribution, which reflects the number of peaks I managed to climb when I tried the three peaks channel for real #nanoporeconf
We are very excited to announce at #nanoporeconf that we have a comparison of R10 and R9.4 data, both native and PCR. We can get better than Q40 genomes on @nanopore R10. Better yet, our data is open, and can be downloaded right now: lomanlab.github.io/mockcommunity/…
Paging @StephenWattam. We should rectify this...
We're seeing modal accuracies over 90% for both the R10 native and PCR.
This is part of our ongoing work on the Zymo community data. These >Q40 genomes are from first binning reads against our draft Zymo reference and assembling with wtdbg2. There's more to come!
We'll be continuing on @Scalene's three peaks protocol and R10.
Now that @scalene revealed to #nanoporeconf that I never made it to Nevis, I feel like we need to finish the last third of our three peaks run.
