This Review discusses the evolving definitions of transcriptional enhancers and the modern experimental tools to identify, characterize and validate them.
The authors discuss how diverse perspectives and methods provide differing but complementary insights into enhancers, each with notable strengths and caveats, and how they might be combined in a comprehensive catalogue of functional enhancers.
The authors propose ‘validated and target-linked’ support as the degree of evidence that should be aimed for in cataloguing non-coding sequences as bona fide human enhancers.
If the evidence falls short of this threshold, as it currently does for nearly all candidate enhancers, the authors suggest strong, moderate and weak tiers to describe candidate enhancers with less or conflicting evidence.
While this Review focuses on enhancers, in particular mammalian enhancers, many of the assays and concepts described are potentially applicable to other classes of non-coding DNA regulatory elements.
