2/ Und nachdem man all die Plaques („Löcher“) auf all seinen zahllosen Platten ausgezählt (und gut dokumentiert!) hat… führt man die Platten ihrer Zweitverwertung zu! 😂

Following up on yesterdays tweet: how to „count“ viruses? The technique is called „plaque assay“ and I’ll share my teaching slide on it below. The gooey medium keeps offspring viruses from swimming around➔ only infect adjacent cells, creating circular holes in the cell layer. 1/

1/ Many discussions on #SARSCoV2 / #COVID19 are quite abstract and focus on infection routes and epidemiology. Further, we often get the question "Can you detect actual #Coronavirus at all, or only 'gene snippets' by this PCR?“ Here come some information, now in English!
2/ The novel #Coronavirus has been isolated countless times from patients, incl. "BavPat1/2020", from one of the first patients in Bavaria, by the team around @c_drosten. Several isolates can be ordered from bio material repositories, e.g. european-virus-archive.com/evag-portal/vi…
3/ The genomes of these isolated and laboratory grown viruses were sequenced, as were thousands of samples from patients around the globe: nextstrain.org/ncov/global. By #sequencing it is possible to clearly identify which exact virus is present. By the way...
1/ Viele Diskussionen um #SARSCoV2 / #COVID19 sind recht abstrakt und drehen sich v.a. um Infektionsrouten und die Epidemiologie. Außerdem immer wieder die Frage „Könnt ihr das #Coronavirus überhaupt nachweisen, oder nur ‘Genschnipsel‘ mit der ‘Drosten-PCR’?“ Hier ein paar Infos!
2/ Das #Coronavirus wurde unzählige mal aus Patienten isoliert, u.a. vom Team um @c_drosten („BavPat1/2020“, aus einem der ersten Patienten in Bayern isoliert). Etliche Isolate kann man (mit entspr. Genehmigung) z.B. beim European Virus Archive bestellen european-virus-archive.com/evag-portal/vi…
3/ Die Genome dieser isolierten und im Labor angezüchteten Viren wurden sequenziert, ebenso wie tausende Proben aus Patienten rund um den Globus: nextstrain.org/ncov/global. Durch #Sequenzierung ist ganz eindeutig nachzuweisen, welches Virus vorliegt. Übrigens…4/
So, nun bezieht auch die Gesellschaft für Virologie #GfV Stellung zum Thema #Schule während der #Corona #COVID__19 #Pandemie. Feste kleine Gruppen, allg. #Maskenpflicht, schelle unbürok. Kurzzeitisolation / Quarantäne von Clustern (=einzelne Klassen) 1/5

Zentrale Prämisse: die Übertragungshäufigkeit von Kindern ist noch unsicher, kann aber nicht als stets sehr gering angenommen werden- man muss mit Ausbrüchen auch im unteren Klassen rechnen. Problem: schwieriger festzustellen, da so häufig symptomlos. 2/5
Darum wichtig: niederschwelliges Testen bei geringstem Verdacht bei Schülern und v.a. auch Lehrern. „Sentinel“-Rolle für Cluster! Ziel: max. 24h bis Befund! Unbürokratische Heimisolation des ges. Klassenverbands (inkl. Lehrer) dann OHNE weitere Tests aber nur für wenige Tage. 3/5
