Can we outsmart #cancer and stop it before it even starts?

Our brand new paper🔥@NatureComms reveals a novel stem-like cell population directly related to #breast tumor initiation.

Let's dig in🧵🧵 Image
First, quick background.

Sadly, everybody reading this knows breast cancer.

It is the most commonly diagnosed cancer in women, with a staggering 1 in every 8 women in the world receiving this diagnosis throughout their lifetimes.
Multiple factors have been shown to modulate breast cancer risk.

You might already know that:

An active lifestyle🏃‍♀️, a good diet 🥦 or breastfeeding 🤱 are protective, while high breast density, radiation exposure or hormone replacement therapy are detrimental. Image

Do night shift duties pose any risks on health?

1. Large number of people are employed in jobs that require working at nights and sleeping in daytime.
Is it healthy? Are there any health hazards associated with this routine?
Let's look at some of the research on this topic.
2. Type 2 #diabetes :
Night shift work, especially rotating shift work including night shifts, is associated with higher type 2 diabetes odds.
Working on #night shifts for >8 nights/month increases the risk of diabetes by 36%. Ref: doi.org/10.2337/dc17-1…

3. Hypertension: In a recent study, night shift workers were found to have a higher #hypertension risk than day shift workers, which increased with increasing frequency of night shift work. Ref: doi.org/10.1016/j.mayo…

Conversations between MP & DBT

MP is in a zoom meeting w #breast radiologist but was interrupted by DBT

DBT: Hey doc, how is it going @RSNA
MP: Hi there, nice of you to interrupt our mtg, was it so important!

DBT:Digital Breast Tomosynthesis
1/
DBT: No doc, but our 2Dmammo were sharing Ur tweets of Chicago yesterday w #Biopsy & me, so I wanted to ask if u are working hard or hardly working
MP: Well “all work no play makes Jack (in my case ‘a Desi’) a dull boy”, so yesterday was fun but today was all work
@aapmHQ
2/
DBT: I heard @DrLizMorris who came to @Hopkins_Rad was a key note speaker today
MP: Hi ‘Rad’ excuse us. Yes, I just returning from her talk. Girl, you would have loved it. She really talked about what Y’all do for breast cancer detection

1/14 🆕 🚨Accelerated article on #breastcancer just out in @nature! #bcsm

We asked:
1️⃣ What drives response to #chemotherapy in #breast #cancer
2️⃣ Can we use #machinelearning to predict response to chemotherapy❓

Link: nature.com/articles/s4158…

A 🧵👇👇👇 @OncoAlert
2/14 We are unable to #predict #response to treatment in clinic…

✅Good response ➡️treatment very effective ➡️better survival
❌Poorer response ➡️treatment less effective ➡️worse survival

Being able to forecast response would be a landmark advance!
3/14 We ran a study in women with #breastcancer @CUH_NHS+@CRUK_CI:
1️⃣who had a #cancer biopsy (which we analysed)
2️⃣received 18 weeks of chemo+/-targeted therapy
3️⃣had surgery (and we measured how much cancer remained)

We associated PRE-therapy profiles↔️POST-therapy response👇

As you may know I’ve a new book out 😊 so for today’s #FollowFriday I’m going to credit everyone on Twitter who helped, inspired or are a source of support listed in #BeingWellInAcademia

It’s going to be a marathon thread! Get ready and prepare to follow some amazing folk 🌟
Let's go! Beginning with a #FollowFriday for @DrHelenKara and @ThomsonPat who are the series editors of the Insider Guides to Success in Academia routledge.com/Insider-Guides… of which #BeingWellInAcademia is a part

Good news! This new human #tumor microbiome study is excellent. They used hundreds of negative controls for contamination + an extensive combo of methods to identify #bacteria in 7 #cancer types (lung, ovary, pancreas, melanoma, bone, brain): science.sciencemag.org/content/368/64… Image
2/ Bacterial LPS + 16SrRNA were frequently detected in all tumor types, w/ #breast cancer harboring a particularly rich and diverse #microbiome. The image below shows LPS/16SrRNA in breast cancer cells. Bacteria were also found in normal breast samples from healthy subjects Image
3/ In one experiment the team grew live #bacteria from three main phyla (Proteobacteria, Firmicutes, Actinobacteria) from fresh breast #tumor samples collected from women undergoing breast surgery

According to the World Health Organization, #breast #cancer is the most common cancer among women worldwide, claiming the lives of hundreds of thousands of women each year and affecting countries at all levels of modernization. A thread 👇
Breast cancer is the most common cancer amongst women in #lka, accounting to 20 per cent of all cancer cases reported in the island. In the case of men, Lip, Oral Cavity and Pharynx cancer is the leading type of cancer and accounts for 20 per cent likewise.
Females have a 100 times higher risk of getting breast cancer than males
(female: male = 100:1). Males when they do get it tend to present unfortunately at an advanced aggressive stage.

Article says 97% of new drugs tested for cancers never make it to market 👉 IMO key to better #cancer therapies will require widespread recognition of the tumor #microbiome/virome...+ the fact that pathogens in such communities can drive changes in #tumor gene expression
Good example of the trend 👉 “Our study demonstrates that #pancreatic adenocarcinoma (PDAC) #microbiome composition, which cross-talks to the gut microbiome, influences the host immune response and natural history of the disease.” cell.com/cell/fulltext/… ImageImage
Or 👉 “...these data indicate that microbial DNA is present in the #breast and that #bacteria or their components may influence the local immune microenvironment. Our findings suggest a previously unrecognized link between dysbiosis and breast cancer”: journals.plos.org/plosone/articl… ImageImage

Ok but how can we expect an analysis like this to produce definitive results without considering the #breast tissue #microbiome 👉 with special emphasis on the #organisms capable of persisting inside many of the cells delineated by the study: frontiersin.org/articles/10.33…
More on the breast tissue microbiome here 👉 “Characterization of human #breast tissue #microbiota from core needle biopsies through the analysis of multi hypervariable 16S-rRNA gene regions”: nature.com/articles/s4159… ImageImageImageImage
Also interrsting 👉 Here, in primates, eating a Mediterranean #diet altered composition of the #breast tissue (mammary gland) microbiome + associated microbial metabolites in the region: cell.com/cell-reports/f… ImageImageImage

“Missing #microbes” are a factor in the development of chronic #inflammatory disease 👉 But personally, I’m more concerned about the fact that many aspects of modern living are driving a rise in “missing #immunity” and the consequent survival of “additional persistent #pathogens
Here’s an example 👉 #Formula feeding is often associated w/ increased chronic #disease incidence☝️ Meanwhile, #breast milk has a #microbiome, but is ALSO filled w/ antimicrobial compounds + metabolites that play a key role in the immune’s system’s ability to manage #infection
This team found that interactions b/t neonatal #saliva and #breast milk released #antimicrobial compounds including H202☝️These compounds inhibited growth of a range of human pathogens: nature.com/articles/s4159… ImageImageImage
