We transcriptionally profiled the unique bdmute phenotype–viable but no stomatal subsidiary cells (SCs)–to identify genes associated with SC formation -> 4th most downregulated gene = BdPOLAR!! /2
CRISPRed bdpolar mutants showed defective SC divisions, much like classical polarity mutants that affect subsidiary mother cell (SMC) polarity factors like PANGLOSS1 homologs. Indeed, also bdpan1 mutants show this defect and double (polar;pan1) has 80% (!) defective SCs /3

Epigenetic “clocks” based on DNA methylation are the most robust and widely used aging biomarker. However, conventional approaches (methylation chip or RRBS) are hundreds of $$ per sample and low throughput.
We wanted to routinely use epigenetic clocks in large studies for low cost. So, we developed Tagmentation-based Indexing for Methylation Sequencing (TIME-Seq) for scalable and inexpensive targeted methylation sequencing of DNA methylation biomarkers.

New #preprint out on mapping effects of DBS for cervical vs. generalized dystonia to the connectome. A brief thread.

Based on N = 80 patients from five DBS centers, we mapped deep brain stimulation sites and looked at whether their structural and functional connectivity profiles could inform clinical outcomes in cervical vs. generalized dystonia.
To derive at a testable hypothesis, we looked at microstructure of the internal pallidum (stimulation site) which is made up from two large processing streams that traverse *orthoginally* from each other. i) the pallidofugal bundle and ii) the pallidothalamic tracts.

We have an exciting new #preprint on @medrxivpreprint ! A novel class of #primaryimmunodeficiency, with the discovery of ITPR3 mutations in two families with combined immunodeficiency. As always, studying #PID teaches us so much about biology! 1/8

The work is based on patients identified @UZLeuven by Rik Schrijvers and @IsabelleMeyts. Both patients had a combined #immunodeficiency with sensitivity to infections, one complicated by peripheral #neuropathy and one by #autoimmune hemolytic anemia. Over to the gene hunters! 2/8
Mutations in ITPR3 were identified by Erika Van Nieuwenhove and Frederik Staels. ITPR3 is part of a #Calcium channel, so we turned to the Serysheva lab @Irina52948708 to predict the impact on structure. Clear as day, the mutations change the charge of the channel. 3/8

First of all, this is an in-silico paper and not an in-vitro paper or a RCT. That itself means that everything written in it is plain speculation. Secondly, the paper is still on a #Preprint server @authorea which means that it has not been peer-reviewed. (3/6)

From >1700 antibodies, we focused in on 50 cross-reactive antibodies that cross-reacted with SARS-CoV and SARS-CoV-2 spike. We then tested the neutralization activity of these mAbs against bat CoVs SHC014 and WIV-1, SARS-CoV, and SARS-CoV-2, and found 4 pan neutralizing mAbs.
We then tested the in vivo activity of these four mAbs against SARS-CoV in mice and found that only one mAb (DH1047) protected against challenge) so we focused on this mAb for more in vivo studies.
