Trials on Vitamin C, E, magnesium, aspirin, calcium

Perhaps, you might be thinking--those are pretty generic interventions 👀

Vit C & E clinical trial was based on concept that there is evidence of oxidative stress in placentas from preeclamptic individuals

OK. Yes, but oxidative stress is a pretty ubiquitous biological event

Excited to see our paper in @JCI_insight! Silver lining of lab shutdown this yr was time to wrap up this project which has been yrs in the making.

We studied a Stox1 KO mouse model to probe mechanisms underlying hypertensive disorders of #pregnancy. 1/

STOX1 is a TF and predicted #preeclampsia risk gene IDed through family-based genetic linkage studies (van Dijk, @NatureGenet 2005). A variant in the DNA-binding domain was proposed to alter protein function ➡️ preE susceptibility. But LOF model had not been studied. 🐁 2/
*We found that loss of Stox1 ➡️ gHTN assd w placental hypoxia + abnormal ECM deposition.
*We observed activation of the uteroplacental renin-angiotensin system in Stox1 KO mice.
*Treatment w an ARB, losartan, rescued the gHTN phenotype, validating the mechanism. 3/
