Antibody landscapes after influenza virus infection or vaccination by Ron Foucier ncbi.nlm.nih.gov/pmc/articles/P…
Much of the global burden of infectious disease today is caused by antigenically variable pathogens, which escape immunity induced by prior infection or vaccination by changing the molecular structure recognized by #antibodies
Human influenza viruses are notorious for their capacity to evolve and evade the adaptive immune response.

Antibody Affinity Maturation to SARS-CoV-2 #Omicron Variants in a #Teachers Cohort medrxiv.org/content/10.110…
Our study, released on @medrxivpreprint this week, looked at the #Covid immunity response of a cohort of primary school staff: 61% had been infected in the 6 months prior to their @Attomarker test and were probably exposed to BA1 or BA.4/5.
The good news is that a number of teachers developed high-quality #antibodies, both in concentration and quality and tuned to the local variant. We think this may have been because they had been repeatedly exposed to SARS-CoV-2.

With passage of the National Alzheimer’s Project Act (NAPA) in January 2011 and the subsequent “National Plan to Address Alzheimer’s Disease”, the war on Alzheimer’s has only just begun. But one key aspect is glaringly absent: specific funding commitments
science.org/doi/10.1126/sc…
More than 5 million Americans now suffer from Alzheimer, and that number is expected to more than double by 2050. Between 1998-2011, there were 101 unsuccessful AD drugs in development and only three approvals (none since 2003)
@AndrewWLo Kenneth @KosikLab
science.org/doi/10.1126/sc…
"If a single drug-development program takes 13 years
from beginning to end and has a 5% probability of success, the expected waiting time for the next approved Alzheimer's disease drug is 260 years"

The lies 2020:

Don’t Panic, we go for #herdimmunity, #children can’t spread it and won’t be harmed

What was really
#wewanttheminfected
And use #schools as a tap for controlled spread. No transparency, no testing no mask

Lets spread the lie that mask simply don’t work

Begin
Here #polarisation starts driven by misinformation and intransparancy by your government. Everybody could see that the measures were nog in line with urgency.

What were they hiding? Why no #zerorisk #zerocovid with borders closed. It could be done

2020: wait we know this will
Lead to many many deaths but the #vaccin is almost here, working hard, we know the #SPIKE is a big thing, hold on #Hopium being built.

Meanwhile lets keep up the lie of children not important to monitor😢 we know from SARS they build less #antibodies lets presume they just not

Gestern kam ein interessantes Interview mit Deutschlands berühmtesten Maßnahmenkritiker, der auch das Schweden-Modell favorisierte. 1/7

2/7
Herdenimmunität = 1 - 1/Ro

Exciting work from @KathyYWei1, @AmeyaHarmalkar & @proteinrosh on predicting scFv developability. TLDR: transformers and CNNs can potentially help prioritise mutations sites for enhancing stability (1/5) #antibodies #machinelearning #proteineng
Context: a single-chain Fv (scFv) is an antibody construct whose heavy and light chains are linked. It's not the conventional "Y" shape molecule, and is useful for engineering / phage display, etc. See @AlissaHummer's post blopig.com/blog/2021/07/a… (2/5)
Thermostability (measured by TS50, the temperature when scFv loses binding) is weakly predicted by 0-shot and fine-tuning via transformers (ESM-1v + ESM-1b). CNNs using sequence and structural (energy) convolutions perform better (?) [hard to tell, sorry!🙈] (3/5)

A "negative" result, but phenomenal thought piece from @naturalantibody / @antibodymap. TLDR predicting antibody-antigen interactions is pretty darn hard (1/5) #antibodies #machinelearning #alphafold

naturalantibody.com/use-case/deepm…
Predicting Ab-Ag interactions is a sub-problem of the protein-protein interaction problem. There are many facets to consider here, including but not limited to, identifying the correct antigen (let alone the correct epitope), the correct paratope, orientation, etc (2/5)
@antibodymap's team show first that true Ab-Ag pairs (i.e. those where we know the Ab binds antigen) and false Ab-Ag pairs (i.e. Ag was randomly given to an Ab), the pIDDT scores are incomparable, suggesting score-based discrimination is HARD. (3/5)

Really excited to announce that AntiBERTa is now published in @Patterns_CP! Here we describe a transformer model that demonstrates understanding of antibody sequences 🧵 (1/6)

#machinelearning #antibodies #drugdiscovery
cell.com/patterns/fullt…
We pre-train a transformer model based on RoBERTa. We exclusively use full-length antibody/B-cell receptor sequences using the MLM objective. Other similar transformers FYI include BioPhi (@prihodad), ABLang (@HegelundOlsen), AntiBERTy (@jeffruffolo) (2/6)
We show that the embeddings pick up nuanced features of BCR/antibody sequences. For example, V gene usage mutational load, and remarkably, B cell provenance. This is all done in a zero-shot setting, i.e. none of these labels were provided during pre-training. (3/6) Image

New #Ebolavirus research shows how 2 powerful antibodies can neutralize #Ebola—and Sudan virus bit.ly/3wg9Lkz

“Finding antibodies with this breadth is important because we don’t know which virus in the genus of ebolaviruses is going to break out next."

1/🧵
To save lives, doctors need broad-spectrum therapies that neutralize as many Ebolavirus species as possible.

This @CellPressNews study shows 2 potent antibodies that can neutralize the well-known Ebola virus and Sudan virus (also responsible for large, deadly outbreaks. Graphical abstract showing ...
The team used #CryoEM to get a clear view of these two antibodies in complex with the #Ebolavirus glycoprotein.

1 antibody, called 1C11, neutralizes the virus by binding with a site on the fusion machinery the virus would normally use to enter and infect host cells. Image

For those wondering how the newest kid on the block, LY-CoV1404 (bebtelovimab) neutralizes variants so well, including #SARSCoV2 #Omicron BA.1.1 and BA.2? The answer is – pure luck! This antibody is like Neo dodging bullets in the matrix…🍀🍀🍀
The binding mode is very similar to imdevimab (REGN10987) but bebtelovimab avoids major imdevimab resistance caused by the N440K Omicron mutation! It also avoids other mutations. Think social distancing of antibodies from mutations...
Can use this 📺 to scroll through and see #Omicron BA.1.1 #SARSCoV2 mutations and where #antibodies bind... Sotrovimab/S309, Casirivimab/REGN10933, Imdevimab/REGN10987, Evusheld (AZD8895/tixa, AZD1061/cilga), Bam (LY-CoV555), Ete (LY-CoV016), and bebtelovimab (LY-CoV1404)

Neutralization data for #omicron is out. By request of @andrew_croxford, I try to explain how scientists investigate #neutralization. First, it's lab-based data. It’s impossible to directly conclude from neutralization data that #vaccines will not work anymore in the field. 1/X
@andrew_croxford Second, assays are legion! I developed one myself, which is different to the one used by the lab of e.g., @CiesekSandra, in terms of #virus, incubation times, etc. Keep that in mind if you compare two papers directly. I will explain how I did my assay. 2/X
@andrew_croxford @CiesekSandra Third, you must understand that such an assay generates data about neutralization of the virus by antibodies in the serum. These assays do not allow for measuring the effect of e.g., T cells or memory B cells.
@sailorrooscout talks about that here: 3/X

As a society we need to
listen to #antivaxxers more
and attack them less.
They are scared.
We don’t know why
until we listen to them.
They want to talk about their fears in the form of statements. People think w words, and often need to process ideas verbally
to sort them.
I just met a very awesome man who has an immune compromised wife.
He is scared of the vaccines.
He kept talking about Malone (quack) and the tech being too new (its not) and all the fear Fear FEAR thats been surrounding the #pandemic and #vaccine rollouts. He is scared.
We DO gain antibodies after natural infection, but they wane. And there aren’t as many of them w natural vs acquired. It takes our bodies a full week to gather troops and launch an attack.
Thats a week of gained if you already have waiting #antibodies.
So how do we communicate?
