1. The @Ligandal antidote to #SARSCoV2 has been getting a lot of attention, but some people are having difficulty understanding how it works and what makes it so promising, so here's a Saturday explainer.
2. There have been many questions about immunity to human coronaviruses and whether vaccines will prompt a long-term protective immune response to #SARS_CoV_2. @edyong209 does a great job of explaining the vagaries of immunity.…
3. @Ligandal opted for an approach that doesn't rely on the human immune system to be effective.

The #SARSCoV2 virus infects human cells via the ACE2 receptor. Here's a computer simulation with the ACE2 receptor in red and the virus in green.

.@NYTimes has done great data journalism on #SARS_CoV_2, filling gaps left by an incompetent US federal response but their recent coverage re: schools is tipping towards sensationalism & divergence from data...…
This coverage is totally freaking people out once again with the idea that preschool-aged children are super-spreaders, which is NOT TRUE! 2/
There’s a lot we don’t know but the overwhelming weight of global data from household, school, & pop-surveillance studies indicates that small children are less likely to contract & spread this virus. Not zero risk but surprisingly low.…

A #SARS_CoV_2 data analysis by modelling.
Five European countries with highest fatalities to date, plus Sweden.

(This updates previous modelling tweeted on July 7th.) ImageImage
Note: This modelling is an analytical tool & not assumption-based predictive type. Minimise assumptions & complexity. Let reality lead the model, not vice versa. Model is compartmental, with additions & mods. Data is used to tests the hypotheses modelled & find parameter values.
for info: An example of previous related analysis from a couple of months ago:

I just want to be clear: while I understand and have similar frustrations with turnaround times for #COVIDー19 testing, I'm afraid the angst is falling on the laboratories...and I do not feel that is fair or appropriate. (Small thread)
The clinical laboratory is full of living, breathing, intelligent human beings. The work takes skill and time. Often, labs are understaffed and there has been a shortage of trained clinical lab scientists FOR A WHILE. There are only so many tests a person can crank out in a day.
Even if a lab has all the staff they need, there's only so much testing that can be done when a) your instruments are full b) you cannot get the supplies you need to run these tests. Seems like many people think labs have unlimited access to very expensive instruments...nope.

1/ Thread: Deeper dive into this fascinating autopsy series from the @davidadorward and colleagues with @COVID_ICECAP @EdinburghUni:

Tissue-Specific Tolerance in Fatal #Covid_19
- N= 11 patients
- Tissue Samples = 37 sites each (23 specific to respiratory tract) → 407 samples
- Mean Age: 77
- 10 (90.9%) patients male
- 4 (36.3%) patients received invasive mechanical ventilation

- #SARS_CoV_2 RNA was detected across ALL sampled organs and tissue sites
- Most frequent = Respiratory tract, GI tract, Heart, muscle
- Less frequent = Liver, Kidney
3/ Organ testing by several methods:

- Multiplex PCR mapped to SARS-CoV_2 genome
--> Presence of S protein evaluated on select PCR positive tissues
- Histological analysis of multiple tissues

.@monaeltahawy has said, FUCK NORMAL, in a short video that rightly pins the pandemic on the status quo that made it possible. Going back to normal isn't what we need. 1/
My friend and colleague @akapczynski and I have talked about a new politics of care that is anything but normal, relies on overturning the political status quo I've lived with all my life. 2/…

Chlorhexidine (CHX) is very much effective against #SARS_CoV_2. The earlier notion that CHX may not work on the 'novel coronavirus' is outdated. Old studies had used very low conc. New evidence shows CHX effective against #2019ncov at 0.05% conc. #COVID19…
Study shows a single rinse with Chlorhexidine (CHX) mouthwash reduces SARS-COV-2 viral load in saliva for 2 hrs. Suggests CHX rinse to prevent #Covid19 spread via droplets.

@profchrisham @ONS There are 4 Pillars. Pillar 1 labs test the Swab by a technique called RT PCR, to see whether the patient has current infection with #SARS_CoV_2. This test is delivered by NHS and PHE labs. Pillar 2 labs do the same but are delivered by the Private Lighthouse labs. 1/n
@profchrisham @ONS 2/n The Pillar 1 labs declare their testing rate, & are a correct number of tests processed. Pillar 2 labs declare the tests processed AND the tests they have sent out. Test sent out should not be counted. Pillar 2 total tests are misleading.…
@profchrisham @ONS 3/n @profchrisham Pillar 3 testing is for antibody tests, to see whether you have had #SARS_CoV_2. These are mostly processed in NHS labs and the numbers are correct. But it is meaningless to add them to the number of RT PCR tests

Excited to announce our new study to develop and validate an inexpensive and sensitive saliva-based method for #SARS_CoV_2 testing in partnership with the @NBA.

Will be led by @awyllie13, @hmkyale, & myself from the @YaleSPH side.…
We'll be using our new #SalivaDirect protocol developed by @VogelsChantal + @DougBrackney. We'll have sensitivity data and a study design overview available on our website soon.…
Our study has 2 aims:

On the lab side, we are preparing #SalivaDirect for FDA EUA, and are validating with many different vendor reagents to drive down costs and not add supply chain bottlenecks. We are not seeking to commercialize any aspect. Our goal is to get this to $1/test

New meta-analyisis estimates: (1) proportion of truly asymptomatic #COVID19 cases and (2) proportion of #SARS_CoV_2 transmission from asymptomatic or pre-symptomatic individuals [1/3]…
Overall, proportion of people who become infected with #SARS_CoV_2 and remain asymptomatic through the course of infection estimated to be 15% [2/3]
40-60% of #SARS_CoV_2 infections are the result of transmission from pre-symptomatic individuals. The contribution from asymptomatic individuals seems to be much smaller estimated at around 6% (but wide confidence intervals). [3/3]

1. It's been frustrating to witness events unfold entirely as some predicted. My heart goes out to all those who've lost loved ones in the #COVID19 pandemic & those who are still suffering with illness.
2. In this March 2nd piece I warned a lethal pandemic was inevitable, the illness would not be mild, immunity would be uncertain, and if we didn't act quickly we'd be trapped with a high infection rate and a broken economy.…
3. We've made all the wrong choices and ended up exactly where I'd feared. So what now? It's clear some governments have given up. Lockdown is too hard. Test, track, isolate is too hard. The economic pain is too severe. They plan to let the virus run through their populations.

1/ THREAD: Memorial Day in a Pandemic

I often struggle with #MemorialDay. With the calls for celebration and happiness on this day. For BBQs and summer fun.

Because nothing about this day is happy.

This day is about #grief. About tragedy. About #death and loss.

🇺🇸 😔 😢
2/ Above all, it is a day of RESPECT. Because Americans gave their lives in service to protect the citizens and country of the United States.

The lives they previously dreamt of were cut short. And that selfless gift of life is one of the highest honors to serve your country. Image
3/ But their husbands and wives will never kiss their spouses again. Their children will never hug their mothers or fathers again. It doesn’t change the visceral grief of parents losing their children to #prematuredeath. #memorialdayweekend2020 Image
